Objective: The increasing degradation of fetal membrane extra-cellular matrix(ECM) has been decribed as the mechanism of premature rupture of the fetal membranes(PROM).Tissue factor parthway inhibitor-2(TFPI-2) was regarded as a matrix-protective function. To sutdy the expression of TFPI-2 mRNA in human fetal membrances, then investigate the role of TFPI-2 in PROM. All of these would provide a new idea for etiology of PROM and the theory basement for clinical therapy.Methods: (1)Fetal membrane samples were prepared from 30 subjects with spontaneous PROM (PROM,n = 9), no labor (cesarean section,CS, n = 11), and labor (normal delivery, ND, n = 10). All patients had no physical and surgical diseases and other pregnancy complication.(2) Messaenger ribonucleic acid expression of TFPI-2 were study with reverse transcriptase polymerase chain recction(RT-PCR).Expression relative density of TFPI-2/β-actin in each group was analyzed with Imagetoo analyzer and compared with F or t test.Result: Eepression of TFPI-2 mRNA in different fetal membrane tissue:(1)in cleft of PROM(0.435±0.059)and in other membrane of PROM(0.454±0.054);(2)in CS(0.545±0.066);(3)in ND(0.517±0.076).(4) The expression of TFPI-2 in PROM was lower than in CS and ND membranes.there was significantly difference(p<0.05). (5)The expression of TFPI-2 in ND was little lower but not significantly different than in CS(P﹥.05).(6) The expression of TFPI-2 in cleft of PROM was little lower but not significantly different than in other membrane of PROM.Conclusion: Our results suggest that The decreased expression of TFPI-2, which can inhibit the degradation of extracellular matrix (ECM) of fetal membrane, could cause increasing of extra-cellular matrix (ECM) degradation and weakening of the membranes lead to rupture.
