Objective:This study was to examine the mRNA expression of HumanMetalloproteinase-9, Cyclooxygenase-2in premature rupture of membranes and normal term pregnancy, and to investigate correlative to the role of them in premature rupture of membrane, as a result it can provide the theoretical basis and new method in prevention and treatment of PROM.Methods:50premature rupture of membranes women who had delivered in the obstetrics of the attached hospital of Guangxi Medical University were selected, and were divided into preterm premature rupture of membranes group(20), and term premature rupture of membranes(30),(30) healthy term-pregnant at the same time were set to in control group. All patients had no physical or surgical diseases and other pregnancy complication. To examine and compare the messenger RNA (mRNA) of MMP-9, COX-2in fetal membranes using real-time reverse transcription polymerase chain reaction, choosing GAPDH as internal reference. The data was statistical treatment with SPSS13.0expressed with mean±standard deviation (χ±s). Results were analyzed with use of Single factor analysis of variance and linear correlation; For these analyses, P<0.05was considered statistically significant. Results:(1) both of the tow groups had expressed MMP-9mRNA and COX-2mRNA;(2) the mRNA expressions of MMP-9and COX-2in fetal membranes were significantly high in women with premature rupture of membranes (included preterm premature rupture of membranes group and term premature rupture of membranes) than in women in controls (P<0.01), however, there were no significantly statistical difference between preterm premature rupture of membranes group and term premature rupture of membranes;(3) the relationship between MMP-9and COX-2in preterm premature rupture of membranes was positively correlated (r=0.738, p<0.05).Conclusion:This study showed that the mRNA expressions of MMP-9and COX-2in fetal membranes were significantly high in premature rupture of membranes group than in controls group, and there was a positively correlation between MMP-9and COX-2in premature rupture of membranes, and suggested that both of them may play a key role in the occurrence of premature rupture in membranes.
