The Mechanism Of Study On The Crude Extract Of Raphanu Affects Gastrointestinal Motility Through The Interstitial Cells Of Cajal

Objective: Raphanus as an edible Chinese officinal herb has been used for a long history and it's benefits on gastrointestinal motility has been confirmed by a vast amount of clinical and laboratory researches.But it's mechanism is still unknow.The investigation observed the interdigestive myoelectric complex(IMC),subtance P(SP),connexin 43(Cx43) and gap junction,which are connect to interstitial cells of cajal (ICCs)change by the crude extract of raphanus (Ecr)to evaluate the possible mechanism of Ecr for exploiting it's pharmacological value on gastrointestinal motility.Methods: Ecr was obtained by using rotary evaporation method.A total of SD rats were randomly divided into A(Ecr) group and B(control)group,1 ml/kg Ecr and 0.9% NaCL were given respectively.After intragastric administration for 1 hour,we observed distributions of ICCs-Cx43 and ICCs-SP in gastric body by fluorescence double-labeling of immunohistochemistry assay,IMC of intestinal by electrophysiological method, and by using the Electron Microscope we observed ultrastructure of ICCs and the change of gap junction.Results:1.The expression of Cx43 and SP which connected with ICCs of Ecr group in myenteric nerve plexus and muscular layer increased obviously in comparison with the control group.Though the expression of SP which lies in mucosa layer and under mucosal layer is not different significant.2.Ecr could improve IMC of intestinal of rats through increasing the intestinal fast waves in unit time3.The ultrastructure of ICCs is similar to each orther except a kind of special gap junction was found in Ecr group.Conclusion: The kinetogenic effect of Ecr is closely correlated to ICCs that be carried out mainly through promoting the electroactivity of ICCs and increasing of Cx43,SP expression which are closely to ICCs.Maybe,it also through enhancing the communitation of ICCs and SMC by the special gap junction.
