| Objective: To investigate the anti-irradiation mechanism of crude extract from pleurotes sapidus, in order to provide the theoretic basis of it for clinical use. Methods: 180 cleaning-grade Kunming mice were randomly divided into 3 groups and treated with 0, 2, and 8 Gy of whole body single X-irradiation, respectively. Crude extract from pleurotes sapidus (3mg/g) were given to the experimental group, while normal saline to the control group. They were killed by dislocation and then the thymus, spleen and peripheral blood samples were taken at 1, 7 and 28 days after irradiation. The numbers of white cell, the thymus and spleen index were detected by routine methods. The expression of bax and bcl-2 were detected by immunohistochemical staining. Results: 24 h after irradiation the expression of Bax protein in thymic and spleenic lymphocytes increased immediately; On the other hand, the expression of Bcl-2 proteins reduced evidently, reaching the lowest level at 24 h. In 2Gy experimental groups, Bcl-2 of pleurotes group is 7.5±1.38; Bcl-2 of control group is 4.4±0.78, one day after irradiation, P<0.05. The expression of Bcl-2 protein was promoted by Crude extract from pleurotes sapidus. Bax of control group is 10.1±1.57, while in pleurotes group it is 6.2±1.34, one day after irradiation, P<0.05. The expression of Bax protein was inhibited. The same significantly difference can be seen 7 days after irradiation, but not significantly 28 days after irradiation.
