Many kinds of aconite root were applied widely in Chinese traditional therapy, with pharmacologic actions, such as stimulating heart, boosting pressure, analgesia and anti-inflammatory action. All these traditional medicines are toxic. The raw extract of crude Sichuan aconite root, the extract of crude kusnezoff monkshood and salted extract of crude lateral root of aconite belonged to poisonous traditional medicine regulated particularly by the administration department. At present, toxicity on aconite root is generally heart toxicity and central nerve toxicity in medical literature.Three kinds of extract (including raw extract of crude Sichuan aconite root, raw extract of crude kusnezoff monkshood and salted extract of crude lateral root of aconite) were evaluated on genotoxcity in this project. The Micronuclei assay in mice, Salmonella reverse mutation assay and Chromosome aberration assay were used for each extract. According to the results, salted extract of strobal was evaluated in Comet assay for the potential of DNA damage.There were three dose groups in every extract in Micronuclei assay of mouse. Gavage doses of the extract of crude Sichuan aconite root were 26.00, 13.00, 6.50 g raw drug /kg. The doses of the extract of kusnezoff monkshood root were 16.60 , 8.30, 4.15 g raw drug /kg. The doses of salted extract of lateral root of aconite were 15.45 , 7.73 , 3.86 g raw drug /kg. Negative control with distilled water and positive control with 40 mg/kg Cyclophosphamide. Gavage one time per day for 4 days.
