Objective:1. To investigate the relationship between the expression of Raf-1 and tumor angiogenesis, and to discuss its clinical significance.2. To establish colon carcinoma transplanted mode in nude mice,investigate the relationship between the expression of Raf-1 and tumor angiogenesis in nude miceMethod:1. Immunohistochemistry technique was used to detect the expression of Raf-1 in 87 specimens of human colon carcinoma, their corresponding adjacent tissues, and incision margins. The patients were from the Department of pathology of Xijing hospital between 2005 and 2007. Microvessel density (MVD) was detected using immunohistochemistry with CD34 labeling. The correlation between Raf-1 expression and MVD with the tumor size, metastasis and differentiation was analyzed. 2. Western blotting technique was used to detect the expression of Raf-1 in different colon carcinoma cells (HT-29,SW480,LS174T). HT-29 and LS174T whose expressions were evidently different, were inoculated into BALB/c nu/nu mice. The growth velocity and weight of transplanted tumors were examined. Lastly, immunohistochemistry was used to detect the expression of Raf-1 and Microvessel density (MVD). The correlation between Raf-1 expression and MVD was analyzed.Result:1. The positive rates of Raf-1 in colon carcinoma tissues, adjacent tissue and incision margins were 86.47% , 37.34%, 11.03%, respectively; there were significant difference between the three values (P<0.01).2. The MVD in three regions were (61.35±11.6), (28.34±6.7), and (8.26±3.7), respectively; And there were significant difference between the three values (P<0.01).3. Western blotting technique was used to detect the expression of Raf-1 in HT-29,SW480,LS174T. Results show that the expression of Raf-1 in LS174T was most ,and HT-29 was least. We choose HT-29 and LS174T to transplant nude mice.4. The growth velocity and weight of LS174T were higher than HT-29 in nude mice colon carcinoma transplanted model.5. Immunohistochemistry results show that the strong positive rates of Raf-1 in LS174T and HT-29 groups were 50%, 10%, respectively; there were significant difference between two values (P<0.01).6. Immunohistochemistry results show the MVD in two groups were (28.32±5.2),(19.23±4.7), respectively; And there were significant difference between two values (P<0.01). Conclusion:1. The expression of Raf-1 in cancer tissues is positively correlated with MVD; and both of them are closely linked with tumor size and metastasis.2. Raf-1 may play an important role in tumor angiogenesis. The expression of Raf-1 is positively correlated with MVD in nude mice colon carcinoma transplanted model.
