For its rarity in the world' species,Metasequoia is regarded by biologists worldwide as the living fossil for biological research.Inspired by the success in the development of Ginkgo flavonoids products,many scholars at home have taken relevant pharmacolotical studies on TFM(total flavonoids of metasequoia),and their researches show that TFM works effectively in anti-arrhythmia and cardiac hypertrophy,and that it will be very useful in curing cerebrovascular diseases if properly developed.In the present thesis,the total flavonoids of Guizhou metasequoia in different periods of the year were measured and a processing scheme for the extraction,separation,purification of TFM was established.Moreover,in this thesis the roles TFM played in anti-oxidation and hypolipidemic were also sudied.The results shown as follows:1.The total flavonoids of metasequoia leaves in Guizhou province varies greatly in different growth period.Samples collected in June have the highest total flavonoid content,and it reaches 1.99-2.15%,which is 30%higher than the data reported in Wuhan.2.The purification of metasequoia would be more desirable if the sample is pretreated under the condition of 70℃(water temperature),30:1(ratio of liquid-solid),and 2h (extraction times).3.After the hot-water purification,the ideal parameters for the ethanol extraction of flavonoids should be:80%ethanol,extraction temperature 70℃,ratio of liquid-solid 25:1,extraction time 0.5h.Under these conditions,the amount of flavonoid extracted is 2.95 times more than that of the counterpart extracted by the traditional methods.4.The optimum conditions for the purification of TFM with D140 macroporous resin should be:the adsorption concentration 2mg/mL,the adsorption velocity 2BV/h. twice circulation of the sample liquid.Elution should be conducted as the following steps:First to dry out the sample liquid with deionized water until colorless.Then the sample liquid should be eluted with 30%and 50%ethanol respectively at the speed of 2BV/h and the two eluates should be abandoned.Finally,the sample liquid is eluted with 3BV 80%ethanol at the speed of 1BV/h.The eluate concerned is collected, decompressed and condensed until its alcohol could not be smelled.Then the refined TEM could be obtained after the sample liquid is freezed and dried in vacuum.With glyptostroboides powder 100 g for a pilot unit,the total flavonoid of the refined TFM amounts to 54.9±1.6%.5.TFM works effectively in eliminating the superoxide anion radicals,hydroxyl radical,DPPH radical.The ratio of elimination,with 100μg/ml TFM sampled,could be 46.3%for O2-·,78.9%for·OH,and 73.1%for DPPH·respecitively.6.TFM plays an important role in hypolipidemic.Research shows compared with the model group the TC,TG,LDL-C of mice serum in different doses group all decreased to some extent.Among them,lipid-lowering effect in medium-dose group and the high-dose group is particularly evident.
