Objective To study the optimum extraction process of three kinds of flavonoids inApocynum venetum flowers, and through the animal experiment, to investigate hypolipidemicantioxidant pharmacological of action on Xinjiang Apocynum venetum flowers extracts forhyperlipidemia model rats, to lay the foundation for further research.Methods (1) Xinjiang Apocynum venetum flowers as raw material, using reflux,flash,ultrasonicthree different extraction process to extract flavonoids of Apocynum venetum flowers, based onthe single factor experiment, design of L9(34) orthogonal test table, to investigate the effects ofdifferent extraction conditions on extraction rate of total flavonoids in Apocynum venetumflowers, To determine the optimal scheme of flavonoids of Apocynum venetum flowers inthe extraction process.(2) The growth adaptability of rats were randomly divided into6groups:The normal group, model group, positive group (simvastatin),Apocynum venetum flowers in thehigh,middle,low dose group,10rats in each group. Except the normal group, the othergroups intragastric administration of high fat emulsion for3weeks to make molds (High fatemulsion made by research group, and the effect verified by the experiment is good), then therats of each group were given corresponding drugs for6weeks, to gain the blood from theabdominal aorta, determining the serum lipids of TC,TG,HDL-C,LDL-C and the antioxidantindex of SOD,MDA,GSH.Results (1) Through the orthogonal experiment, the best craft of extraction for reflux of Apocy-num venetum flowers is: Time is50min, ethanol concentration is50%, ratio of solid to liquid is1:40,2times of extraction; The best craft of extraction for flash-type of Apocynum venetum f-lowers is: Ratio of solid to liquid is1:55,ethanol concentration is50%, time is130s,2times ofextraction; The best craft of extraction for ultrasound of Apocynum venetum flowers is: Ratioof solid to liquid is1:45, ethanol concentration is45%, time is55min,2times of extraction.(2)Blood fat index: Compared with the normal group, TC, TG, LDL-C, LDL-C/HDL-C levels ofthe rats in the model group significantlincreased, the change has significant difference (P<0.01),HDL-C content decreased, the change has significant difference (P<0.01).Compared with the model group, positive group, Apocynum venetum flowers in the middle, high dose group can sig-nificantly decrease the level of TC (P<0.01or P<0.05); Positive group, Apocynum venetum flow-ers in the high,middle,low dose group can significantly decrease the level of TG (P<0.01or P <0.05); Positive group, Apocynum venetum flowers in the high,middle,low dose group can signi-ficantly increase the level of HDL-C (P<0.01or P<0.05); Positive group, Apocynum venetum fl-owers in the high,middle,low dose group have a significant reduction of LDL-C (P<0.01or P<0.05); Positive group, Apocynum venetum flowers in the high,middle,low dose group have a si-gnificant reduction of LDL-C/HDL-C (P<0.01or P<0.05); Apocynum venetum flowers in the lo-w dose group has no significant difference in reducing the level of TC (P>0.05). The antioxidantindex: Compared with the normal group, the SOD and GSH of serum of the rats in the model gr-oup significantly decrease, the changes have significant difference (P<0.01);The MDA content si-gnificantly increased, the change has a significant difference (P<0.01). Compared with the modelgroup, the serum SOD of rats of positive group, Apocynum venetum flowers in the low, middle, high dose group increased significantly, the change has a significant difference (P<0.01or P<0.05);the serum MDA of rats of positive group, Apocynum venetum flowers in the low, middle, high dose group decreased significantly, the change has a significant difference (P<0.01or P<0.05); the serum GSH of rats of positive group, Apocynum venetum flowers in the low, high dose group increased significantly, the change has a significant difference (P<0.01or P<0.05); The level ofGSH of Apocynum venetum flowers in the middle dose group has no significant difference compared with the model group (P>0.05).Conclusion (1) The three extraction methods of reflux extraction, flash extraction and ultrasonicextraction can extract flavonoids from Apocynum venetum flowers, and the method isstable, repeatability conditions is good. Flash extraction is fast and safe, don’t destroythe medicinal ingredient, the extraction rate is high.(2) Positive group, Apocynum venetumflowers in the low, middle, high dose group have a certain extent to the therapeutic effecton hyperlipemia rats, the high dose group has a better therapeutic effect, the improvement of thesituation of each index is better than middle and low dose group, and improvement of thesituation of some index is better than positive group, so it is worthy of further research anddevelopment.
