Objective: To explore the effects of glucolipid metabolism of compound preparation of Inonotus obliquus, Redix pueraria and Rhizoma Polygonati etc.on experimental diabetic rats and probe the function of protection and rehab on damnification of pancreatic island and it's mechanism. In the meantime, provide scientific theoretics and experimental evidence for clinic which could cure diabetic.Methods: The rats were divided into 6 groups: normal control, diabetic model, positive control (glucophage 200mg/kg.d) and low density compound preparation, middle density compound preparation, high density compound preparation. The rats had been tested for 40 days. We should measure fasting blood glucose (FBG) of every groups before the experiment and after. We measured the level of glycosylated hemoglobin (GHb), total cholesterol (TC), triglyceride (TG), low density lipoprotein (LDL-C), superoxide dismutase (SOD), malonal dehyde (MDA), alanine transaminase (ALT) and aspartate transaminase (AST) after the experiment. We executed them by the method of neck mutilation and took the pancreatic island, hepatic and nephridial tissue from their abdominal cavities. Then we made use of HE staining method, PAS staining method and immunohistochemistry staining method and observed pathological changes.Results:The results of biochemical measurement showed that compare compound preparation of Inonotus obliquus, Redix pueraria and Rhizoma Polygonati etc group and diabetic model group, the former was able to degrade the levels of blood of FBG, GHb and the content of TC, TG, LDL-C, MDA, significantly. It could advance SOD activity and degrade ALT and AST activity. All exhibited concentration dependent relationship. The results of pathomorphology changes of pancreas, liver and nephros is that compare compound preparation of Inonotus obliquus, Redix pueraria and Rhizoma Polygonati etc group and diabetic model group, the former made the number and bulk of pancreatic island increase, the number of B cell in pancreatic island and insulin-secreting in B cell increased, the content of glycogen particle in hepatic cell increase remarkably, the thickening of glomerular basement membrane (GBM) and the hyperplasy of intercapillary cells were not remarkably and decrease the positive things of mesangialregion.Conclusions: Compound preparation of Inonotus obliquus, Redix pueraria and RhizoaPolygonati etc. had effects of decreasing blood glucose, regulating blood fat,antioxidation cleaning free radical, improving the function of impaived liver, and thefunction of protection and rehab on damnification of pancreatic island, liver andnephros. The mechanism is indistinct, might be concerned with complex action ofmultiple active components from compound preparation and function of reparation andamelioration of B cell of pancreatic island.
