[Objective]To explore the hypoglycemic effects of Inonotus Obliquus of type 2 diabetic rats made by two methods.[Methods]1(1)Health male SD rats were randomly divided into normal control group and diabetes mellitus model group.Type 2 diabetic rats model were made by two ways which were feeding fat emulsion and injecting Streptozotocin as well as feeding fat diet and injecting Streptozotocin in diabetes mellitus model group.(2)We chose the first method to establish type 2 diabetic rat model eventually by comparing the effects to the content of free fatty acid,total cholesterol and triglyceride in serum of rats made by two different methods.Dividing the successful models into 4 groups randomly according to their blood glucose level:model group(normal saline 10ml/kg),Inonotus Obliquus low-dose group(1000mg/kg),Inonotus Obliquus high-dose group(2000mg/kg),and positive control group(Metformin Hydrochloride 900mg/kg).(3)After 8 weeks of medicine,fasting but water available for 12 hours,ether anesthesia,drawing out all the blood in their hearts.Separating blood serum and measuring fasting blood glucose and the content of total cholesterol,triglyceride and free fatty acid in serum.Removed pancreatic tissues of rats in different groups,and observing the morphology of pancreatic B cells after hematoxylin-eosin staining under the microscope.[Results](1)Compared with normal control group,the content of free fatty acid in serum of rats increased after feeding fat diet for 4 weeks(P<0.05)and the content offree fatty acid in serum of rats increased obviously after feeding fat emulsion for 15 days(P<0.01);Compared with fat diet model group,the content of free fatty acid of rats in serum was different from it in fat emulsion model group(P<0.05);(2)Compared with normal control group,the content of total cholesterol of rats in serum in fat diet model group and fat emulsion model group increased obviously(P<0.01);Compared with fat diet model group,the content of total cholesterol of rats in serum is obviously different from it in fat emulsion model group(P<0.01);(3)Compared with normal control group,the content of triglyceride of rats in serum is different from it in fat emulsion model group(P<0.05),and the content of triglyceride of rats in serum in fat diet model group increased obviously(P<0.01);Compared with fat diet model group,the variance of the content of triglyceride of rats in serum is relatively small;(4)Compared with normal control group,the content of fasting blood glucose of rats in serum increased obviously(P<0.01)after injecting Streptozotocin for 1 week in type 2 diabetic rats models made by feeding fat diet and feeding fat emulsion;The content of fasting blood glucose of rats in serum decreased in fat diet model group,while the content of fasting blood glucose of rats in serum increased in fat emulsion model group after injecting Streptozotocin for 7 weeks;What's more,the content of fasting blood glucose of rats in serum is different from fat diet model group and fat emulsion model group(P<0.05);(5)Compared with normal control group,the content of free fatty acid in serum of rats in diabetes mellitus model group increased obviously after feeding fat emulsion for 8 weeks(P<0.01);(6)Compared with normal control group,the content of fasting blood glucose in serum of rats in diabetes mellitus model group increased obviously after feeding fat emulsion and injecting Streptozotocin for 8 weeks(P<0.01);(7)Compared with normal control group,the content of fasting blood glucose in serum of rats in model group increased obviously(P<0.01);Compared with model group,the content of fasting blood glucose of rats in Inonotus Obliquus high-dose group(2000mg/kg)decreased obviously(P<0.01);(8)Compared with normal control group,the content of total cholesterol and triglyceride in serum of rats in model group increased obviously(P<0.01);The content of total cholesterol of rats was different between Inonotus Obliquus low-dose group(1000mg/kg)and model group(P<0.05)and the content of total cholesterol in serum of rats in Inonotus Obliquus high-dose group(2000mg/kg)decreased obviously(P<0.01)compared with model group;And compared with model group,the content of triglyceride in serum of rats in Inonotus Obliquus low-dose group(1 000mg/kg)and Inonotus Obliquus high-dose group(2000mg/kg)decreased obviously(P<0.01);(9)Compared with normal control group,the content of free fatty acid in serum of rats in model group increased obviously(P<0.01);And compared with model group,the content of free fatty acid of rats in Inonotus obliquus high-dose group(2000mg/kg)was different(P<0.05);(10)Compared with normal control group,the structure of islet is not complete in model group,and the cells are dispersed;Compared with model group,the structure of islet in Inonotus Obliquus low-dose group(1 000mg/kg)and Inonotus Obliquus high-dose group(2000mg/kg)are almost complete,and the amount of islet B cells increased.[Conclusionl(1)The model made method of feeding fat emulsion and injecting Streptozotocin is better than feeding fat diet and injecting Streptozotocin;(2)Inonotus Obliquus can decrease the content of free fatty acid in serum of type 2 diabetic rats that made by feeding fat emulsion and injecting Streptozotocin and can decrease the content of fasting blood glucose?total cholesterol and triglyceride obviously in serum;(3)Inonotus Obliquus can repair the damaged islet cells.
