Objective: To establish the rapid and stable model of the heterotopic heart transplantation in rats and improved it.Methods: To make a model with Wistar rats (200-250g) which based on the models designed by Ono and partly of the procedures were improved. All models were divided into 2 groups in a random. Group A (n=40): Firstly, prepare donor, then cut heart and afford cold ischemia. Secondly, prepare receptor and then perform abdominal heterotopic heart transplantation. Group B (n=40): Prepare receptor and donor, after cut heart and directly perform abdominal heterotopic heart transplantation. To observe the Re-beating time and the pulsation of the transplanted heart. Rats of successful operation were phlebotomized in inferior vena cava in the third day. Measuring the contents of IL-2,IL-4 in rat serum with ABC-ELISA method. Simultaneity took out two groups of successful heart to do pathology.Results: Established and improved successfully of model abdominal heterotopic heart transplantation in rats. Group A: The hot ischemia time was (1.5±0.5) min. The clod ischemia time was (30.5±0.5) min. The transplanted heart re-beat in 5~10min. Operation achievement ratio was 70%. Group B: The hot ischemia time was (1.5±0.5) min. The cold ischemia time was (15.5±0.5) min. The transplanted heart re-beat in 4min. Operation achievement ratio was 80%. The contents of IL-4 maintained at a relatively low level in rat serum of each group. The contents of IL-2 remained at a relatively high level. The pathological examination of transplanted heart found that: myocardial cells swelling, necrosis, interstitial hemorrhage, edema, vascular endothelial rupture, and vascular thrombosis.Conclusion: To compare two methods of the operation, in the group B, the time of operation and ischemia is shorten, the whole process simply and can be completed by a single man. The recovery was quickly after operation. The operational achievement ratio of group B is higher than group A.
