Heterotopic Heart Transplantation Model In Rat

【Objective】:To establish a quick and effective heterotopic heart transplantation model in rat.【Methods】:Male SD rats served as donors and Wistar as recipients,the ascending aorta of allograft was anastomosed to the recipient abdominal aorta,The recipient underwent a left nephrectomy,and The pulmonary of donor heart connects with the receptor left renal vein by homemade's polyethy lene pipe directly.Form the blood route:receptor abdominal aorta—donor arteriae aorta—aeteria coronaria—cardiac muscle—vena coronaria—right atrium—right ventricle—arteria pulmonalis—receptor left renal vein—inferior vena cava.After heart transplantation,touching abdomen daily to observe the transplanted heart.That transplants surviving more than 24 hours marks success.【Result】:1.The model of heterotopic heart transplantation has been sucessfully established,the mean ischemic time was(37.8±1.8)min in 35 experiments.The success fulrate was 91.4%.Complications were anastomotic bleeding,thrombosis.2.In donor myocardium,swelling necrosis,hypertrophy and breaking myocardial fibers were seen by routine light microscopy.There was an increase and hypertrophy of interstitial cells and most of which were lymphocytes.Other organs had significant changes.Mitochondria of myocardium were markedly swollen,with dissolution of cristae in donor hearts.In some mitochondria,the matrix space was dilated and vacuolated mitochondria formed.【Conclusion】:This method has advantage of simpleness,and requiring no operating microscope.So it is easily to be spreaded.
