The Study On The Effects Of ShuGan JianPi JieDu Recipe On The Lymph Node Matastasis Of Liver Cancer And The Expression Of MMP-9, VEGF

Objective:To observe the ShuGan JianPi JieDu(SGJPJD) recipe to the H22 model of liver cancer cells in mice invasion and the impact of lymphatic metastasis,and further explore its mechanism,with a view to effective clinical use of the SGJPJD recipe detoxification of liver cancer,especially cancer the transfer of advanced treatment provide an objective basis.Method:SPF-48 male mice,weighing 18±2g.To establishe the tumor lymphatic metastasis model.Later,mice were randomly divided into 6 groups:Control group,CTX group, Middle-dose of SGJPJD,High-dose of SGJPJD,Middle-dose of SGJPJD with CTX,High-dose of SGJPJD with CTX.The next day,the chemotherapy drug was given by intraperitoneal injection for 4 days,traditional Chinese medicine for 21 d gavage,the model group to the contour saline.The day after the end of the experiments,the mice were killed by cervical dislocation,we weighed the tumor,the spleen,the thymus and the side of popliteal,iliac artery next to the door and renal lymph nodes.Calculated the rate of inhibitory effect of liver carcinoma and the index of the lymph nodes,the spleen,the thymus.HE stain,light microscopy level of metastasis.We measured MMP-9 expression of the mice through Immunohistochemis -try technique method and real-time fluorescence quantitative Reverse Transcription-Polymerase Chain Reaction(RT-PCR) to test the expression of VEGF mRNA.To explore the mechanism of the SGJPJD recipe on the liver cancer of the lymphatic transfer mechanism.Results:It has showed out that the SGJPJD recipe has remarkably suppressing effects of mice H22 liver cancer and lymph.It can significantly increase the inhibition rate of CTX to transplanted mice H22 liver cancer and lymph(compared with the control group,P＜0.05);In addition,the SGJPJD recipe can significantly improve the mice's immunologic function, especially for High dose of the SGJPJD recipe(compared with the control group,P＜0.05).The SGJPJD recipe can decrease the expression of VEGF mRNA and MMP-9 protein expression (compared with the control group,P＜0.05),and the SGJPJD recipe with CTX group which compared with the CTX group and model group could have statistic differences(P＜0.05).Conclusion:The SGJPJD recipe can inhibit liver cancer in mice H22 and mice lymph node metastasis,and enhance effects of chemotherapy-therapy.Their role may be related to reduce VEGF,MMP-9 expression;The SGJPJD recipe can also enviously enhance the immunologic function of H22 liver cancer mice.
