Objective:To observe the JianPi ShuGan JieDu(JPSGJD) recipe on the expression of p21 and K-ras in the provocative liver cancer model of rat by DEN and the stagnation of liver-QI with deficiency of the spleen stimulating factor,explore its preventive and therapeutic effect and the mechanism,with a view to effective clinical use of the JPSGJD recipe detoxification of liver cancer,especially providing an experimental basis.Methods:SPF-72 SD male rats,six weeks old,weighing 100-120g. The rats were randomly divided into six groups:To establishe the DEN Control group,Low-dose of Control group, High-dose of Control group;Model group, High-dose of Model group,Low-dose of Model group. And then,the control groups were offering 95μg/l DEN dilution interruptablly; Except that,the model groups were extra acceptted various ways of PIXUGANYU,such as drenching rhubarb, narrowing action and so on. High-dose of all groups were given medicine according to 30mg/kg;Low-dose of all groups were given medicine according to 10mg/kg;Control group and Model group were given physiological saline,and all of them were medicated administrtion team for 150 days gavage.During the experiment,we observed the activities of rats,after that killed,we weighed the liver,the spleen and the weight of rats.Calculating the rats of inhibirory effect of liver carcinoma and the index of the spleen.HE stain,observing the liver cancer through light microscopy, We measured p21,K-ras expression of the rat through Immunohistochemistry technique method.Results:At the beginning, the DEN Control group had no clear difference between each other.Through light microscopy,we found that pathological change of the DEN Control group was most serious in all control groups.with the experiment going on,the rats in the Control group decreased the drink and food, and lost weight,moved slowly,,their pelage were neat. The rats in Low dose of control group were slightly normal,but their eating were reduced,weight losing,and the pleges were unnormal. The High-dose of control group was common in every direction. The DEN Control group of liver pathological changes was obvious:the surfaces of the liver were scobinate,gloomy,and many small tubercles diffusing;But the surface of the High-dose of control group liver changed insidiously which having thick granules,but no typical cancerous node.The pathology change of the Control group is the most serious.The Control group's orgnization of lobuli hepatis are distroyed and displaced by nothogolial, hyperpla--stic fibrous tissue divided virgin lobuli hepatis,and circumvolutio inequality of size hepatic cell,the central veins absence can be seen,a little chromatospherite are incement,and the quantities ate increased obviously, inflammatory cell infiltrations in portal area are increased slightly,fibrosis which are penim the lobuli hepatis are more obviously.The spleen index of the High-dose of Control group is increased most remarkble compared with the Control group and the Low-dose of Control group(P<0.01),which between the statistical significance also is;On the contrary,the liver index of the Control group is increas-ed most clearly in these three groups (P<0.01). There is statistical significance of the p21 and K-ras expression in three control groups, the p21 expression of High-dose and Low-dose of Control group increase more remarkble,but K-ras is in contrast. The model rats were enterorrhea after being drenched rhubarb,and the splenic asthania,stagnation of liver-QI,they decreased the drink and food, and became irascible. with the experiment going on,they beggan loseing weight and hair,staying together, moving slowly,losing temper and so on.The unitary condition of High-dose and Low-dose of Model group were better than Model group.The surfaces of the Model group liver were rough,gloomy,and many small ashy or yellow tubercles diffusing that the number of High-dose and Low-dose of Model group were less obviously.The pathological change of Model group is most serious:the hepatic cells in Model group are confused,not equirotal, anomaly and partly had vacuole, necrosis,partly the volume of the hepatic cells became larger than before and atypical. the cellular nucleus are big,some hepatic cell is big and a heteromorphism,the blood vessel hyperplasia is aggravate.The weight of High-dose and Low-dose of model group is obviously higher than Model group (P<0.05); The spleen index of the High-dose of group is increased most remarkble compared with the Model group (P<0.01); The liver index of Model group is increased more obviously than other Model groups(P<0.01); The expression of p21 and K-ras of High-dose and Low-dose of Model group increase more remarkble compared with Model group,but K-ras is in contrast (P<0.01)Conclusion:The JPSGJD recipe can remarkably improve immunologic function of rats. Its conspicuous prenentive and theapeutic effect may be related to increase the expression of p21 and inhibit the devitalization of K-ras.
