Effects Of Quchiling On Cholinergic System In Dementia Rats Model Induced By Ibotenic Acid

ObjectiveTo investigate the effects of quchiling on brain cholinergic function in rats suffered from Alzheimer's Disease.Methods(1) By inject ibotenic acid to the Meynert nucleus in basal forebrain made rats AD model. After recovering one week,the therapy groups oral administration quchiling with concentration of(11.3g·kg^-1·d,5.67 g·kg^-1·d,2.83 g·kg^-1·d) for 28 days,the masculine drug was Huperzine A.The morris water maze test,analysis of AchE,Ach,ChAT activites,the pathological observation of the Meynert nucleus in basal forebrain were carried out to evaluate the effects of quchiling on the AD model rats.Results(1) ethology:compared with sham operation group,delitescence of model group prolonged obviously(P＜0.05),middle and high dose group of quchiling compared with model group, delitescence shortened(P＜0.05),compare with model group,the spatial probe ability improved(P＜0.05).Compare with model group,the withdrawal reflex ability no significant diffenrece.(2)biochemical indicator:high dose group of quchiling compared with model group, activity of AchE upgraded(P＜0.01),middle and high dose group of quchiling compared with model group,activity of ChAT upgraded(P＜0.05),compare with model group,contents of Ach no significant difference.(3)morphology change:compared with model group,AchE positive fiber density increased (P＜0.01). ConclusionThe suitable dose of quchiling can improve the AD model rats' learning and memory deficiency,enhance the activity levels of ChAT and AchE,increase the density of AchE positive fibers in their brains,it has the antagnonism effect to the impairment of cholinergic neurons, recover their function.
