Effect Of Bushen Huoxue Representative Prescription Yizhi Jiannao Granule On AD Rats' Cognition And Memory Ability And The Expression Of ChAT And TrkA In Hippocampus CA1

OBJECTIVE To observe Bushen Huoxue prescription Yizhi Jiannao granule's effects on AD rats' cognition and memory ability and the expression of ChAT and TrkA in hippocampus CA1 and to explore the possible mechanism.METHODS 40 male SD rats were randomly divided into 4 groups by weight such as normal group(group A), sham-operation group(group B), model group(group C) and treatment group(group D). Then the last two groups were injected by Aβ_25-35 in both sides of hippocampus CA1 to make AD models. Rats of group B were injected by the same volume of ddH₂O in the same way. No treatments were done to group A. Rats of group D were given Yizhi Jiannao granule while the rest groups were given ddH₂O for 4 weeks . All the groups were tested with Morris water maze test(MWM) after medical treatment. Finally decollated all the rats to be dead and collected the specimen to observe the expression of ChAT and TrkA by RT-PCR method.RESULTS1. Compared to group C, group D's average incubation period shortened and the platform jumping times increased in MWM (P<0.05). Compared to the group A, group C's average incubation period prolonged and the platform jumping times reduced significantly(P<0.01).2. Compared to group C, group D's expression of ChAT increased in hippocampus CA1(P<0.01). Compared to the group A, group C's expression of ChAT decreased in hippocampus CA1(P<0.05).3. Compared to group C, group D's expression of TrkA increased in hippocampus CA1(P<0.01). Compared to the group A, group C's expression of TrkA decreased in hippocampus CA1(P<0.05).CONCLUSION1. By injecting condensed Aβ_25-35 to both sides of rats' hippocampus CA1 can make representative AD model rats.2. Bushen Huoxue prescription Yizhi Jiannao granule can remarkably improve AD rats' space cognition and memory ability and increase the expression of ChAT and TrkA in AD rats' hippocampus CA1 .The curative effect is affirmative.3. The potential mechanisms of Bushen Huoxue method's curative effect on AD may relate to decreasing the absence of neurons, protecting the function of neurons and improving the function of cholilergic system and etc.
