Background and Objective:As phosphodiesterase (PDE)â…¢inhibitor, milrinone has both inotropic and vasodilator actions to improve hemodynamics in patients with congestive heart failure, but some findings indicated that despite its beneficial hemodynamic action, long-term therapy with milrinone increased the morbidity and mortality of patients. Olprinone, a newly developed inhibitor of PDEâ…¢, improve cardiac performance not only through its inotropic effects, but also through its vasodilating effects . We performed this study to assess the efficacy and the security of olprinone versus milrinone for the congestive heart failure.Mthodse:Totally 32 patients with severe chronic heart failure(NYHAâ…¢-â…£) to treatment with olprinone (n=15) or matching milrinone (n=17). End points were clinical outcome, echocardiographic left ventricular ejection fraction, left ventricular dimensions, left ventricular volume, left ventricular shortening fraction, and sternite as well.Results:After one week treatment we observed that The administration of olprinone and milrinone to the same degree on congestive heart failure, P>0.05. The results of therapy were assessed as follows: the olprinone effective rate 86.7% versus milrinone 82.4% by NYHA curative effect score; the Boston effective rate of olprinone was 60% and milrinone got 62.5% as well. The total adverse effect of olprinone was only 1 while 4 of the administration of milrinone, but there were no statistics difference. Further studies were needed to determine the adverse effect of both drugs.Conclusion:The results of this study suggest that olprinone significantly improved cardiological function as same as milrinone. There were no statistics difference on adverse effect between two drugs.
