The Effect Of Clopidogrel On P-selectin And CD40L Derived From Platelets And Neutrophil From BAL Of Mice

Objective To observe the effect of Clopidogrel on P-selectin and CD40L derived from platelets and neutrophil from BAL (Bronchial alveo lavage) in the asthmatic model mice.Methods Female BABL/c mice were devided into 3 teams with random: the control team,the asthmatic team and the treatment team by Clopidogrel, to detect the positive rates on expression of P-selectin and CD40L of platelets by the Flow cytometer(FCM), and to detect pathological and cytological changes.Results The p-selectin positive rates of platelets expressed in the conrol team,asthmatic team and treatment team(1.23±1.02)%,(4.52±1.45)%,(1. 42±1.12)%, respectively, the P-selectin positive rates of platelets expressed in conrol team and asthmatic team,asthmatic team and treatment team show significant difference(P<0.05);The P-selectin of platelets positive rates expressed after activated by thrombin in the conrol team,asthmatic team and treatment team are (45.12±4.03)%,(44.68±5.34) %,(44.23±5.42) %, respectively, show no difference(P>0.05); The CD40L positive rates of platelets expressed in the conrol team,asthmatic team and treatment team (2.43±1.22)%,(5.63±1.52.) %,(2.24±1.33) %,respectively, the CD40L positive rates of platelets expressed in conrol team and asthmatic team,asthmatic team and treatment team show significant difference (P<0.05); The CD40L of platelets positive rates expressed after activated by thrombin in the conrol team,asthmatic team and treatment team are(22.45±3.98)%,(23.18±4.04)%,(23.23±3.51)%, respectively, show no difference(P>0.05); The rates of leukocyte of bronchial alveo lavage(BAL) in conrol team,asthmatic team and treatment team (6.64±1.86),(25.60±2.66),(7.23±2.02)%,respectively, The rates of leukocyte of bronchial alveo lavage(BAL) in the asthmatic team and conrol team,in the asthmatic team and treatment team show significant difference (P<0.05),in conrol team and treatment team show no difference (P>0.05);Expression of P-selectin and CD40L show significant positive correlation.Conclusion There is platelets activation during the asthmatic attack mice; CD40L can be a marker of platelet activation.clopidogrel can reduce the inflammation action during the asthmatic attack, ADP can be a most difficult factor inducing the platelets activation.
