| Objective:To investigate expression of SPARC protein in normal ovarian tissues,benign ovarian epithelial tumors, borderline malignancy,ovarian epithelial carcinoma and to study its relationship with pathologyical behavior of ovarian epithelial carcinoma .Methods:To select 127 cases of patients who were treated surgically at the Affiliated Hospital of Qingdao University Medical College from September 2004 to June 2006. All cases include 13 cases of metastatic ovarian epithelial carcinoma,63 cases of primary ovarian epithelial carcinoma which were divided into several groups according to pathological type ,clinical stage and histological differentiation,and 9 cases of ovarian epithelial borderline tumor, 20 cases of ovarian benign epithelial tumor,22 cases of normal ovarian tissue. All the patients of ovarian neoplasma did not receive any adjuvant radiotherapy ,chemotherapy and immunotherapy before surgery. The differential protein expression of SPARC in normal ovarian tissues and ovarian epithelial carcinoma was analyzed by tissue microarray technology and immunohistochemistry analysis and located using well characterized monoclonal antibodies. The Semiquantitative data were analyzed by t-test, which were analyzed by SPSS11.5 and the significant size of test of this study was validated as value of 0. 05.Results: The expression of SPARC protein was significantly down-regulated in ovarian epithelial borderline tumor and ovarian epithelial carcinoma compared to ovarian benign epithelial tumor and normal ovarian tissue (P<0.05). It was not associated with the tissue type,lymph node metastasis (P>0.05) but the histological grade,ascites,caul metastasis and the clinical stages of ovarian epithelial carcinoma (P<0.05). SPARC located in cytoplasma except 16 cases.Conclusions: These findings indicate that SPARC is marked and may play an inhibited role in the development,growth and metastasis of ovarian epithelial carcinoma. The expression of SPARC protein, together with other indexes,may be a valuable marker for diagnosis,detection and prognosis of ovarian epithelial carcinoma . |
