Objective:Pathological scar includes hypertrophic Scar and keloid,the pathogenesis of which has not been clear.At present,the scholars in domestic and foreign generally believe that the pathological Scars is formed with excessive proliferation of fibroblast and accumulation of excessive collagen and extracellular matrix.Which is a substantial interest in Pathogenesis.This experiment is to investigate the difference of SPARC in Pathological Scar and normal skin.Explore the biological contribution of SPARC in the formative course of hypertrophic Scar and keloid.Methods:(1)All these 63 patient scars(32 hypertrophic scars(hyperplasia group),31 keloids (scar group )) and 10 cases of normal skin tissue taken as control (control group) were applied.The method of immunohistochemical SP were used to detected three groups of tissue expression of SPARC. (2)Fibroblasts culture from primary cells: Pathological scar and normal skin were harvested to culture fibroblasts according to the document method, cell passaged after fusing. The fourth to sixth generation cells were collected to detected the difference between Pathological scar and normal skin , which used Western blot method.Results:1,Through Western blot method detected the expression of SPARC and found Pathological scar and normal skin all expressing SPARC.2,Made use of gel Image Processing system analysis Pathological scar Gsa/β-action has difference with normal skin Gsa/β-action.The former Gsa/β-action bigger than latter.Date through statistics analyses,make use of t test,(P0. 05).Conclusion:1,Pathological scar and normal skin are all express SPARC which demonstrates that SPARC Involved in the development of Pathological scar and normal skin.2,The proliferation of collagen fibers, smooth muscle cells, skin appendage, infiltration of inflammatory cells and microvascular endothelial cell, in Pathological scar ,may be related to SPARC expression3,The expression of SPARC,which in pathological scar tissue fibroblasts ,was significantly higher than that in normal skin fibroblasts(Western blotting test results)...
