Objective:To investigate the expression of secreted protein acidic and rich in cysteine(SPARC) in cervical carcinoma and its relationship with biological behavior of cervical carcinoma. Methods:To select 63 cases of patients who were treated surgically at the Affiliated H-ospital of Qingdao University Medical College from January 2004 to July 2006.All cases include 59 cases of squamous cell carcinoma and 4 cases of adenocarcinoma. All the patients of cervical carcinoma did not receive any adjuvant radio- therapy, chemotherapy and immunotherapy before surgery.67 cases of cervical intraepithelial neoplasia, and 23 cases of normal cervical tissue. The differential protein expression of SPARC in normal cervical tissues, cervical intraepithelial neoplasia(CIN) and ovarian epithelial carcinoma was analyzed by tissue microarray technology and immunohistochemistry analysis and located using well characterized monoclonal antibodies. The semiquantitative data were analyzed by t-test,which were analyzed by SPSS11.5 and the significant size of test of this study was validated as value of 0.05.Results:The SPARC was expressed differently in all cervical carcinoma,CIN and normal cervical tissues, and its expression was measurably higher in cervical carcinoma than in control groups(P<0.05).The expression of SPARC was associated with the histological grade, interstitial infiltration(P<0.05),not associated with the tissue type, lymph node metastasis, clinical stage, age, recurrence, tumor size(P>0.05).Conclusions:SPARC might play a role in the development, growth and metastasis of cervical carcinoma.SPARC maybe a possible biomarker of squamous intraepithelial lesions(SILs) and invasive cervical cancer.
