Immunoglobulin is a kind of glucoprotein bearing antibody activity,which mainly reside in blood plasma,also in body fluid,tissue and secretory juice. immunoglobulin in human plasma can be divided into five categories,those are immunoglobulin G(IgG),immunoglobulin A(IgA),immunoglobulin M(IgM), immunoglobulin M(IgM),immunoglobulin D(IgD),immunoglobulin E(IgE). IgG is the main immunoglobulin among those kinds.Immunoglobulin in hen blood transferred to fowl ovi vitellus can provide protection during development process of hen.,which is called immunoglobulin of fowl ovi vitellus(IgY).IgG is the main immunoglobulin in fowl ovi vitellus.IgM and IgA are mainly exsiting in serum protein.IgY belong to polyclonal antibody which has many advantages on disease's diagnosis and prevention.Usually polyclonal antibody is quite difficult for large-scale preparation and multiple preparation can induce bad homogenicity.The hen can obtain specific antibody from ovi vitellus continually after immune response..IgY with high purity and high specificity,can not make cross sero-reaction and anaphylactic response like other mammal.So it is the safest biological agent,which can be in mass production substituting the antibody prepared by mammals,such as immuned mouse,cavy or rabbit and etc.Recently the investigation and application on IgY is the hot spot and very athletic topic in the field of immunology.Meticillin sodium resisting Staphylococcus aureus is an important pathogenic bacteria causing hospital onset of infection.It has high pathogenicity which is drug fast to many antibacterials and therapeutically rather troublesome.Taken Meticillin sodium resisting Staphylococcus aureus as antigen immuned hen,anti-meticillin sodium resisting Staphylococcus aureus IgY is prepared.Anti-meticillin sodium resisting Staphylococcus aureus IgY is invesgated on physico-chemical property and biological activity,which establish the foundation for diagnosis and prevention in the future.[Objective]To observe the yield,purity,source and Stability of the anti-meticillin sodium resisting Staphylococcus aureus antibody(MRSA).。[Methods]Taken the anti meticillin sodium resisting Staphylococcus aureus antibody(MRSA) as antigen immuned Leghorn fowl,IgY was extracted by improved water dilution method.Antibody's content was determined by double UV optical wavelength method,Antibody's purity was detected by SDS-PAGE electrophoresis.Antibody's source was determined by Western blot.IgY's constancy to temperature,acidity or alkalinity,pepsase and trypsase was determinated by ELISA.[Results]Antibody contained 13.3 mg/mL of yolk liquid,purity of antibody attained 96%,immunoblotting proved that IgY and IgG in chicken serum had the same molecular wieght and antigenicity,IgY had favourable heat stability and tolerance to acid,base,pepsase and trypsase. [Conclusion]High yield of IgY which had highly purified specificity and favourable biologic activity can be attained by Improved water dilution method.
