Hypericin,with important pharmacological action of Antidepressants, anti-tumor and anti-virus,was selected in this dissertation to be the research object。First,the best synthesis conditions of the hypericin molecular imprinted polymers was calculated.Second,deep and systematical researches had done to develop new isolation method.At the same time,It had been built a method to determine hypericin in the extraction of Hypericam perforatum L..Details are as follow:1.The pre-organization system of molecularly imprinted polymer with hypericin as template,Acrylamide as functional monomer,ethylene glycol dimethacrylate as crosslinking agent was studied by PM6 method in MOPAC 2007.The influence of concentration of functional monomer and crosslinking agent on the concentration of template-monomer complex wad investigated by computer simulation.The optimized synthesis condition of hypericin molecularly imprinted polymer was also studied theoretically.It is helpful to interpret experiment phenomena and synthesize molecularly imprinted polymer with high affinity and selectivity.It has provided the theoretical conduction in hypericin molecularly imprinted polymer.2.MIT was developed with an increasing speed recently.In order to expand the application of this technology in natural products,a molecular imprinted polymer(MIP) was prepared by a molecular imprinting technique using hypericin as the template molecular at the first time.At the conduction of the pre-organization system of molecularly imprinted polymer,the molecular imprinted polymer was investigated in equilibrium binding experiments to evaluate its molecular recognition,binding characteristics.The results showed that,compared with non-imprinted polymer,the imprinted polymer exhibited much higher affinity for hyperforcin from complicated samples(Hypericam perforatum L. extracts).Application the imprinted polymer to separation of hypericin from complicated samples showed the promising future of MIT in traditional Chinese medicine area.Anymore,through the methods of FT-IR spectra,MIP was proved to be a multi-porous polymer,which could be a explanation of its high affinity to template molecular.3.A method has been built to determine hypercin in Hypericam perforatum L.with TLC Scanning.The sample was separated on silica(G) with solvent petroleum ether/EtOAc/CH3OH(1:4:2),the silica G is treated by the Na2CO3 solvent(0.4 mol/L).The CS-930 scanning densitometer of Shimadzu was selected.The detection wavelength was 591 nm.The results showed that the calibration curves were linear in the range of 0.0~14.4μg for hypericin(r=0.9892).The range of recoveries were 96.4 %~100.1%.The method is rapid,accurate and reliable.And the stability of hypericin in the condition of different pH,temperature and light were discussed.In this paper,using computer applications for molecular imprinted polymer preparation conditions of the theoretical analysis,data analysis was provided for the condition of MIP synthesis.This can help to reduce the number of experiments,to save the human,material and financial resources.Furthermore,TLC was a simple,rapid and accurate detective method which provides for the study of the Molecular imprinted polymer.The application of MIP to the isolation of hypericin in complicated samples provided theory and practice base and also a new route for the separation and purification of effective components in natural products.
