The extract of hypericum perforatum is the botanical drug to sell well besides ginkgo leaf in the international market. Hypericin is the most bioactive component in hypericum perforum, which has the pharmaco-activity of antivirus, antitumor, antidepressant, antibiosis and dephlogisticate. The content of hypericin is seldom in hypericum perforatum. RP-HPLC is used to separate and purificate hypericin from plants in foreign country. The extraction technology of hypericin was developed lately in our country. At present, the most of production technology are still concentrating on the extraction. The method of separation and purification about hypericin is little. Thus, it is significant to apply the new type medium to separate and purificate the hypericin.The technology of separating and purifying hypericin were reaserched by monodisperse microspheres of PEK and PDCX in this thesis. Main research is as follow:A great quantity of tannins in the extract of hypericum perforatum induces the trouble for following separation and purification. Studying the technology of getting rid of tannins in extract of hypericum perforatum in this text. The technology of getting rid of tannins as follows: 6 times-weigh volume ethanol was used to dissolve the extract of hypericum perforatum with ultrasonic for 30 minutes, then, to get rid of tannins with the method of improved gelatin precipitation. Tannins were got rid completely. The recovery and purity was 85.3% and 0.27%.The hypercin is unstable and its content is seldom in plants, which cause the inconvenience in hypericin manufacture. The stability of hypericin under the conditions of light and temperature and ascorbic acid were studied by HPLC in the text. The conclusion as follows:the light and temperature are outstanding factor for the stability of hypericin. So, hypericin should be avoided the light and high temperature in producing and preserving.Study on the purification technology of hypericin with the microspheres of PEK and PDCX and ODS in the text. The performances of static adsorption and desorption, dynamic adsorption and desorption of PEK microspheres were studied. The purification technology of PEK as follows: 1 bed volume of the 0.6 mg/mL concentration extracts was add in the chromato bar, then, water and gradient concentration ethanol solutions of 10%, 30%, 40%, 60%were used to elute for 1 bed volume, 1 bed volume, 1.5 bed volumes, 1.5 bed volumes, 4 bed volumes separately. Hypericin were mainly concentrated on the 60% ethanol. The recovery and purity were 89.9% and 3.2%. The first pure production was purified with PDCX microspheres. The purification technology of PDCX as follows: 1/3 bed volume of the 0.45 mg/mL concentration solution was add in the chromato bar, the eluant were gradient acetone. The purity of the second pure production was 33.0%. The second pure production was purified by ODS, the production purity can be up to 91.6%.
