Objective Investigating the viability and insulin level of rat islets through the action caused by Pomegranate peels extract(PPE). Methods PPE was extracted with 95% ethanol then obtained the ethanol extract. In addition the PPE was extracted with series of organic solvents and obtained the different kinds of extracts. Then the following experiments have been done on this extracts. The islet cells were divided into common control group, Gilemal masculine control group, different PPE groups were cultured for different days respectively,then to evaluate the viability of islet cells with the method of MTT, and select the extract which can promote the growth of islet cells, then the experiment were carried out as follows. (1)The islet cells of PPE groups and control group were cultured with high glucose(25.2mmol/L) DMEM for different days respectively and the supernate was harvested regularly to determine the concentration of insulin by radioimmunoassay.(2)The islet cells of common control group, high glucose group and high glucose+ PPE groups were cultured for 72 hours and the supernate was harvested to investigate the levels of insulin secretion by the glucose-stimulating insulin releasing test.(3)The islet cells of common control group,palmitic acid group,palmitic acid + PPE group,olein acid group and olein acid + PEE group were cultured for 36 hours, then the supernate was harvested to investigate the levels of insulin secretion by the glucose-stimulating insulin releasing test. Results (1)Pomegranate Extract of Ethylacet- ate (PEE) can promote the growth of islet cells ,the viability of islet cells was markedly higher than that of control group (P<0.01). (2)The insulin level of PEE groups was obviously higher than that of control group (P<0.01) .(3)The insulin levels of high glucose + PEE group, palmitic acid + PEE group and olein acid + PEE group were significantly higher than those of high glucose group (P<0.01),palmitic acid group and plein acid group respectively,while those insulin levels were markedly decrease as compared with those of control group (P<0.01). Conclusions PEE not only contribute to improve the viability of islet cells and increase the function for insulin secretion, but also could protect the function of islet cells.
