Objective To investigate the change of Inhibin B(INH-B)level in the serum and follicular fluid of patients with polycystic ovary synthrome (PCOS) , clucidate correlation between Inhibin B and BMI and other five sexy hormones, as well as the relationship with PCOS.Methods 1. Serum study: Serum was collected from 45 patients with PCOS and another 20 control women with regular manses in the day 3 of the menstrual cycle. Serum inhin B concentration was assayed by enzyme-linked immunosorbent assays. Serum LH, FSH, T, E2, PRL concentrations were also assayed at the same time. Correlation analysis was applied to serum inhibin B and sex hormone, BMI.2. Follicular fluid study: inhibin B and T were assayed in the follicular fluid of≤10mm follicles from 11 PCOS patients and size-matched follicles from 6 normally cycling women. Peripheral serum were collected and assayed at the same time. Comparasion and correlation analysis of follicular fluid and peripheral serum hormone levels were also conducted.Results 1. In the serum study, mean serum inhibin B level was markedly increased in the PCOS group. The level of INH-B in the serum of nonobese patients with PCOS was higher than that of obese patients with PCOS and all the controls. Simple regression analysis shower that in PCOS,serum inhibin B level was positively related to LH, FSH, T, but the same results were not seen in the controls. Negative correlation was found between INH-B and BMI both in patients with PCOS and controls.2. In both control and PCOS ovaries, follicular fluid concentrations of inhibin B, T were much higher than those found in serum. There was significant and positive correltion between follicular fluid level of inhibin B and T (P<0.05) both in PCOS and controls.Conclusions In women with PCOS, serum and follicular fluid inhibin B concentrations were higher than that found in control women. Inhibin B was higher in follicular fluid than in serum level, intrafollicular and serum inhibin B were significantly correlated to T concentrations. Serum inhibin B was significantly correlated to LH, FSH and T concentrations. These data support the concept that PCOS is associated with a dysfunction in the endocrine and autocrine and/or paracrine regulation. Obesity has an independent effect on inhibin B concentration. Inhibin B may take part in the formulation of dysregulation of steroidogenesis in PCOS. Testing and regulating the level of serum INH-B is value to promote the treatment.
