【Objective】To study the effect of poplyglycoside of tripterygium wilfordii hook.f. on inflammatory cellular factors of peritoneal elicited macrophage activated by lipopolysccharide in mice, for discuss control drug or measure to the hyperecytokinemia in excessus inflammation response.【Methods】The peritoneal elicited macrophage were separated, purified and activated by LPS in mice, then cultured in vitro with a series concentration of TWP, at the same time the macrophages were activated by LPS. Test the level of cytotoxicity and phagocytosis through MTT colorimetry. The TNF-αactivity and IL-6 level of cellular supernatants were determined by MTT colorimetric assay and ELISA respectively.【Results】The level of cytotoxicity and phagocytosis of MФcan be inhibited significantly (p<0.01) by TWP (6.25~100μg/L)during 4~24h, and the inhibitory effect is correlated with the dosage and time. The activity of TNF-αand the level of Il-6 in MФ, could be significantly inhibited (P<0.01) by TWP (6.25~100μg/L and 6.25~50μg/L, respectively) during 4-24 h and 12h respectively in a time and dosage dependent manner.Conclusion: TWP can inhibit the TNF-αactivity and Il-6 level on macrophage activated by LPS.
