Objective To study the effect of oral contraceptives(OCs) antagonizing tripterygium wilfordii polyglycoside(TWP)-induced gonad toxicity in the female SD rats.Methods40female SD rats wered ivided randomly into four groups,each group has10rats:TWP group,OCs group, TWP with OCs group, and blank control group.Obtain vaginal exfoliated cells smears and observe the changes of estrous cycle everyday after10weeks medication.Then kill the rats after3weeks and take celiac artery blood for testing follicle stimulating hormone(FSH),luteinizing hormone(LH),estradiol(E2) by ABC-ELISA,compare the weight of uterus and ovary and the number of follicle,observe the histomorphology of uterus and ovary with microscope.Results Compared with the blank control group and TWP with OCs group,oestrous cycle of TWP group is incomplete or extended;E2was (40.4174±4.9372)pg/ml,obviously lower than the rest of the three groups,FSH was (1.0634±0.1539)mIU/ml,LH was (5.9467±1.0360)mIU/ml,obviously higher than the rest of the three groups(P<0.05);the weight of uterus and ovary were (0.0487±0.0097)%and (0.1470±0.0047)%,the thickness of endometrium and myometrium were (57.20±1.81)um and (24.40±1.84)um,obviously lower than the rest of the three groups(P<0.05);the number of growing follicle and graaffian follicle were (22.50±2.55) and (2.00±1.89),obviously under than the rest of the three groups(P<0.05),but primordial follicle were (36.50±2.64),compared with the rest of the three groups(P>0.05).Conclusion Diane-35can decrease TWP-induced gonad damage in the female SD rats and plays a potential role in protecting ovarian function.
