| 80mg tiamcinolone acetonide(TA) periocularly injected, then research the drug metabolic process.Methods: The right eyes received a single periocular injection of 1ml TA suspension as experimental group; likewise the left eyes received 1ml PBS as control group. 20 New-Zealand rabbits were used and divided into five subgroups. In five subgroup intraocular pressure (IOP) and the dark adapted electroretinograms(ERGs) were recorded before and after injection; then immediately obtained retinal tissues for light and electron microscopy and procured aqueous humor and vitreous for drug concentration measure by spectrophotometer.Results:IOP and ERGs had no significant difference between experimental and control group. Tissue structure had no abnormality by microscopy. Aqueous drug concentration was maximal at 2 week(547.6±118.0ug/ml) and gradually descended and presumably disappeared at 8 week. Vitreous drug concentration was 35.91±14.97ug/ml at 1 week and gradually descended to 11.54±5.37ug/ml at 7 week. At the same time, it had significant linear correlation with time and presumably lasted 11 weeks.Conclusion:Periocular injection of 80mg TA is safe for IOP and retinal structure and is effective for ocular drug concentration. |
