Objective Analyzes the concentrations of triamcinolone acetonide following a single sub-Tenoninjection or periocular injection in retina and vitreous of rabbits to provide the experimentalbasis for clinical applications.Methods: Sixty healthy adult gray rabbits divided into two groups randomly. One groupreceived sub-Tenon injection of TA (40mg in0.4ml) in their right eyes. The other groupreceived periocular injection of TA (40mg in0.4ml).And the fellow eyes don’t received anythingas the blank control eyes. Three animals were killed at each designated time points (1day,3days,7days,14days and28days) and The intraocular pressure (IOP) and slit-lamp examination wererecorded before injection and was executed. And the globes were snap frozen and dissected intovitreous and retina. The concentrations of TA in the vitreous and retina were analysed using highperformance liquid chromatography (HPLC) and the statisticas.Results: The retina regression equation is Y=0.604X+0.0691(R2=0.9997, P<0.05).The vitreousregression equation is Y=0.6258X+0.0372(R2=0.9998, P<0.05). They are presenting a goodlinear correlation.The posterior sub-tenon group: TA concentration were (1.555±0.654) g.g-1(0.729±0.375) g·mL-1in retina and vitreous at post-injection day1. The concentration of TA inretina and vitreous achieve maximum at post-injection day3and were (11.850±4.427) g.g-1,(1.621±0.539) g·mL-1. After that the concentration showed a declining trend. The perioculargroup: TA concentration were (0.157±0.058g.g-1(0.050±0.027g·mL-1in retina andvitreous at post-injection day1. The concentration of TA in retina and vitreous achievemaximum at post-injection day3and were (1.040±0.292g.g-1,(0.143±0.070g·mL-1.After that the concentration showed a declining trend. No any ocular complication was foundthroughout the studying duration. No significant alteration of IOP was found after injection ofTA or normal saline.Conclusion:1. After a posterior sub-tenon or periocular injection of TA in rabbit eyes. the concentrations oftriamcinolone acetonide can be found and maintain a certain time.2. In this experiment research condition, compare with the periocular group, the posteriorsub-tenon group can get higher concentration in the retina and vitreous organization of rabbiteyes, and both concentration has significant differences.3. The effective drug concentration can be retained in the vitreous cavity and retina after aposterior sub-tenon or periocular injection of TA in rabbit eyes.
