The study of proteomics is a main orientation of life science after the research of genomics.As structure and function of protein influence the change of vital movement directly .The research of gene in the transcriptional level only reflect the change of gene expression product in finite level,but the formation of the protein which bring into full play must process many steps and regulations,include post-transcriptional processingtranslation regulation, posttranslational processing and so on. So only studying protein direcly can explain all kinds of vital phenomena.At present,the means of research need a words of development .so look for protein analytical technique which utility and shotcut is a most significant component element. The emergence of the techology of protein array brings a new means to the research of proteomics .A main domain of the proteomics research is the research the protein expression level in different kinds of pathologic status.Thus the antibody array which is miniaturization,integrattion,high-flux is a very good reseach tool.It`s also a array that developed fastest in all protein arrays ,and it`s mature increasingly.Some of these antibody arrays had been used in clinical application, however most of them applied in scientifiec research domains .The reseach of signal transduction is a hot spot in base investigation,this project is an corporation between our lab and McGill University, Montreal, QC, Canada.We are mainly in charge of antibody preparation for antibody array,the work includes construction of the plasmid,protein expresson,protein purification,antibody preparation and antibody purification.In the study we mainly prepare the protein that correlates with signal transduction ,such as PAK1,RhoA,Rac1,MEK1,Rho-GAP,SH2(2)-NCK,SH2(2)-PLC, C-SRC and so on( see the appendix ).Firstly we have prepared the highly titer,highly specific yolk antibody--GB1-VFT,and we also need a secondary antibody of highly titer high specificity and stablely.So we also have prepared a monoclonal antobody against Fc fregment of the IgYand labeled the monoclonal antibody with HRP.We found the initial plateform of antibody array–antibody platform .include chicken yolk antibody platform,mouse monoclonal antibody platform.It privides a very important tool for the continue research of antibody array and proteomics .At the same time the GB1-VFT yolk antibody is a good help for the research of GPCR.,We also meet some difficulity in preparing yolk antibody ,it is problem that we should solve next step.
