Echinops grijisii Hance(Compositae) is widespread in southeastern China.The roots of this plant has been recorded in Chinese Pharmacopoeia(1995 Edition) as Radix Echinopsis.Radix Echinopsis is a traditional Chinese medicine used as clear heat,expel miasma and stimulate milk secretion.From the 95%ethanol extract of the roots of Echinops grijisii Hance,22 compounds were isolated by means of solvent partition,repeated normal and reversed phase column chromatography on silica gel and Pre-HPLC.Based on spectroscopic analysis(NMR,UV,IR and MS) and physical-chemical characters,their structures were identified as Thiophenes:α-terthienyl(1),5-(but-3-en-1-ynyl)-2,2'-bithiophene (2),isocardopatine(3),5-(4-isovaleroyloxybut-1-ynyl)-2,2'-bithiophene(4),2-(3, 4-diacetoxybut-1-ynyl)-5-(prop-1-ynyl) thiophene(5),5-(3,4-diacetoxybut-1-ynyl)-2, 2'-bithiophene(6),5-(penta-1,3-diynyl)-2-(3,4-diacetoxy-but-1-ynyl)-thiophene(7), 5-(3-acetoxy-4-isovaleroyloxybut-1-ynyl)-2,2'-bithiophene(8),5-[5'"-(propyne-1)-2'" -(3"-acetylene-6"-(but-1,3-diynyl)-cyclobutane)]thiophene-2,2'-bithiophene(9), 5-(4-isovaleroyloxy-butyryl-1)-2,2'-bithiophene(10),5-(4-hydroxybut-1-ynyl)-2,2'-bithiophene (11),2-(penta-1,3-diynyl)-5-(4-hydroxybut-1-ynyl)thiophene(12),5-(3, 4-dihydroxybut-1-ynyl)-2,2'-bithiophene(13),2-(prop-1-ynyl)-5-(5,6-dihydroxy-penta-1, 3-diynyl)thiophene(14),arctinol-b(15),2-(penta-1,3-diynyl)-5-(3, 4-dihydroxybut-1-ynyl)thiophene(16),2-(prop-1-ynyl)-5-(3,4-dihydroxybut-1-ynyl) thiophene(17);Phenolic compounds:2-methoxy-4-methylbenzaldehyde(18),3-(3, 4-dimethylphenyl)acrylaldehyde(19);Fatty acids:9,12-octadecadienoic acid(20); Hydrocarbons:(Z,Z,Z)-3,6,9-heptadecatriene(21);Phenylpropanoids: chlorogenic acid(22).Among them,compounds 9,10 are new compounds;compound 21 is a new natural product;compounds 7,12,13,15,18,19,21,22 were firstly isolated from this plant.Different fractions of the 95%ethanol extract were examined for their antitumor activities using K562 and HL60 human myelogenous leukemia cell lines,and the antitumor parts were mainly found in the dichloromethane extract.Meanwhile,the antitumor activity of the dichloromethane fraction was also studied in vivo in S180 implanted ICR mice.By MTT and SRB methods,the in vitro cytotoxic activities of the thiophenes isolated were assayed based on different human cell lines.
