A great many plants contain the monomer of flavonoid- These components have more than one hydroxy radical(R.OH),and provided with antioxidant effect of hydrogen radical(H.),can scavenge the super oxide anions(O2),hydroxy free radical(OH.) and other free radical activity.Objective:The purpose of this study was to investigate the possible mechanisms of extractives of the Chinese Herbs of flavonoid in carbon tetrachloride -induced acute hepatotoxicity and testis oxidative damage in rats.Methods:60 male SD rats were randomly divided into groups.Except the normal group,other rats received a single dose of CCl4(2 ml/kg in corn oil,intraperitoneally). Three hours after CCl4 intoxication,flavonoid drug group and the positive drug rats received either the Chinese Herbs of flavonoid(200 mg/kg) or bifendate(100 mg/kg) by gastrogavage twice a day for one.CCl4-induced liver damage was shown by significant elevation of serum aminotransferase levels,and liver,testis tissues were obtained for the contents detection of hepatic MDA,SOD,GSH-Px.The liver and testis pathological change were evaluated under light microscope after HE staining. At the same time,hepatocyte P450-2E1 mRNA and iNOS mRNA expression were detected with Semiquantitative RT-PCR analysis.Results:In contrast,rats given both the Chinese Herbs of flavonoid and bifendate supplement had less elevation of serum aminotransferase concentrations associated with less severe lobular damage of hepatocytes than rats receiving CCI4 alone(P<0.05, P<0.01),but contents of ALB hadn't obviously increasing.Additionally,compared with the model group,the contents of MDA decreased remarkably in flavonoid groups liver and testis tissues(P<0.05).SOD,GSH-Px had significant elevation of liver tissues(P<0.05),but didn't changed remarkably in testis.The results of HE staining,the flavonoid groups could reduce CCl4 intoxication and protect the tissues.Moreover,the Chinese Herbs of flavonoid treatment inhabited the elevation of hepatocyte iNOS mRNA expression,at the same time, increased the hepatocyte P450-2E1 mRNA expression.The protective effects of flavonoid group surpass the positive group.Conclusion:Extractive flavonoid of the Chinese herbs has protective effects on acute hepatic injury and antioxidative damage of rats induced by CCl4.
