The Observation Of Therapeutic Effects Of Compound Traditional Chinese Medicine On Hepatic Fibrosis

ObjectiveTo investigate therapeutic effects of compound traditional Chinese medicine on hepatic fibrosis in rat modle and discuss its mechanisms.MethodsThe rat models of hepatic fibrosis were induced by carbon tetrachloride and interfered with different doses of compound traditional Chinese medicine or Shosaikoto.Compound Biejiaruangan Tablet was used as positive control.Serum levels of ALT,AST,ALB,HA,LN,PC-â…¢,TGF-β1,TNF-α,and levels of Hyp, MDA,and the SOD in liver tissue were detected;Pathologic examinations of liver sections were performed by routine HE and Masson staining,Grade of hepatic fibrosis were evaluated according to SSS system.Immunohistochemical method was executed for detecting collagen alphal(â… ),(â…¢) and PDGF-BB in liver tissue.Activated HSC was detected byα-SMA immuohistochemistry stain.HSC apoptosis was detected by terminal deoxynucleotidyl transferase(TdT) dUTP nickend labeling(TUNEL) and Desmin immuohistochemistry stain.ResultsCompared with the fibrotic group,serum levels of ALT,AST,HA,LN,PC-â…¢,TGF-β₁,TNF-α,Hyp,MDA decreased obviously(P＜0.05),Compound traditional Chinese medicine could increase the level of SOD in hepatic tissues obviously(P＜0.05),which was depressed in fibrotic group;liver pathological variations of fibrotic was improved obviously in groups interfered with compound traditional Chinese medicine,and the score of hepatic fibrosis in compound traditional Chinese medicine group reduced significantly(P＜0.05).Compound traditional Chinese medicine treatment could reduced expressions of collagen alphal(â… ),(â…¢) and PDGF-BB(P＜0.05).Aactivated HSC increased significantly in fibrotic group compared with that of normal group,and few apoptotic HSC were detected.However,less activated HSC and more apoptotic HSC were detected in compound traditional Chinese medicine treated group compared with those of fibrotic group.Conclusions1.Compound traditional Chinese medicine could improve hepatic functions and liver pathological variations and heal hepatic fibrosis.2.Anti-peroxidation.,enhancing degradation of collagen in liver tissue and reducing the expression of cytokine may be the functional mechani sms of compound traditional Chinese medicine against hepatic fibrosis3.Inhibiting HSC activation and proliferation,and inducing HSC apoptosis may be another functional mechanisms of compound traditional Chinese medicine against hepatic fibrosis...
