| Hepatic fibrosis is regarded as a pathologically repair reaction followed various kinds of chronic liver disease.Fibrosis mainly presents the imbalance of the ECM compound production and degradation,which makes the ECM deposit pervadingly in liver.Hepatic fibrosis is the necessary pathologic phase of chronic liver injury from a variety of cause including virus(HBV,HCV),hemadsorption , alcohol abuse,autoimmune,drug induced, cholestatic and metabolic disease. With the advance of fibrosis, nodule formation and altered hepatic function reselt to cirrhosis at the end stge,and hepatic cirrhosis can seriously harms patient's physical and mental health.Hepatic fibrosis is a kind of common disease that is associated with a significant morbidity and mortality,which ia a major worldwide health care burden.For it is the common pathological feature in the development of chronic liver diseases,the key to control these diseases would be preventing,treating and reverding hepatic fibrosis.Many studies proved that liver fibrosis can be reversed. At present,it is the deficiency of medicince specificly treating HF, such as Colchicina which is already reported can inhibit HF, but it has a large poisonal adverse reaction. But in the recent years, the experimental and clinical studies on anti-hepatic fibrosis of Chinese traditional medicine have achieved remarkable development. HF belongs to the category of chest and hypochondrium, jaundice and meteorism of Chinese medicine. It caused by loemia of humid heat or insobriety leading to humid heat,which are detain, sorrowful emotion and so on, leads gradually to disorder the fuctions of liver, spleen and kidney, to make QI-blood ,body fluid and its metabolism fight each other, liver nervure blocked. The key is blood stasis caused by humid heat for a long time. The complex prescription of chinese materia medica is a experienced prespcription and consisted of ten kinds of Chinese herbal,All of them together have the effect of clearing heat and detoxicating,promoting blood to resolve stagnation.Ecperimental and clinic researchs have showed that the compound have better effect to treat chronic liver diseases hepatic fibrosis. According to it, by animal experiment combinated with the theories of modern medicine, we approach initially the therapeutic effect of this complex prescription which is against HF and provide experiment evidences of clinical medicine.Objective: This study is designed to investigate the therapeutic effects of traditional Chinese medicine on experimental fibrosis in rats and explore its related mechanism.Methods :Hepatic-fibrosis models were prepared by the means of subcutaneous injection of carbon tetrachloride preparation with 10weeks, wister rats were divided randomly into 4 groups: control group, model group, complex prescription of chinese materia medica group,complex prescription of Danshen group.Drugs were given on the ten weeks after models prepared. In normal control group and model control group, the rats gave intragastric administration with 10ml/kg distill water of identical volumetric every day which successively administrated eight weeks. In masc control group, the rats gave intragastric administration with 100ml/kg salvia miltirorrhiza dripping pills of complex prescription every day, In complex prescription of chinese medicine, the rats gave intragastric administration with suspension of 10g/kg crude drug power every day which successively administrated once a day for eight weeks. During this time , the rats fed with the standard forages, drinked tap-water in 20～22℃of room temperature. After the last administrated, rats were kept without food but water, 12h later, blood was taken from abdominal aorta, and the serum was separated.measured the levelof ALT, AST,ALB,MDA.A small piece of liver tissue was taken, fixed in the 10% formal liquid;another small part of liver tissue taken was washed by normal saline, made to be 10% liver homogenate, then measured HYP, the activity of the HA,the laminin(LN),precollagen typeⅢ(PCⅢ),collagen typeⅣ(CIV)was evaluated according to radioimmunoassay method.Result:1.The result of histopathology:we can get the result from microscopic examination:normal control group:the structure of the liver lobule was clear; there was no obviously desmoplasia.model group:we can see some rugosity in the surface of liver ,the structure of liver lobule has been destroyed,and many pseudo-lobule which were divided by crudeness or fineness fibrous connective tissue appeared. Masculine medicine group: compared to the model group ,the degree of patho-injury in the rat hepatic tissue relieved obviously,in this group,there was four rats which hepatic tissue imoroved obviously and the separation of fibrous connective tissue, four rats'hepatic tissue were improved,but we still could see small amounts separation of fibrous connective tissue,only one rat hepatic tissue sepatation widen obviously . Complex prescription of Chinese crude drug group: there was four rats'hepatic tissue imoroved obviously and the separation of fibrous connective tissue, five rats'hepatic tissue were improved,but we still could see small amounts separation of fibrous connective tissue.we could make a conclusion:this complex prescription of Chinese crude drug has a notable affect to anti-rat hepatic fibrosis. 2.Hepatic functional parameter:compared with contol group,hepatic function of rat in model group was significantly increased with respect to ALT,AST. hepatic function of rat in model group was significantly decreased with respect to ALB(P<0.001). Complex Prescription group: AST,ALT decreased obviously, and had obvious difference compared to the model group(p<0.01), ALB obviously increased compared to the model group(p<0.05).3.the effect of MDA in serum:compared with contol group,the MDA in serum of rat in model group was significantly increased(P<0.001).compared with model group ,the MDA in serum of rat in Complex Prescription group increased obviously,and has signifince in statistics.4.Serum hepatic fibris index: compared with contol group,the LN,HA,C-Ⅳ,PC-Ⅲin serum of rat in model group was significantly increased(P<0.001). compared with model group ,the LN,HA in serum of rat in Complex Prescription group decreased obviously(P<0.01), C-Ⅳ,PC-Ⅲwas also decreased compared to model group, and has signifince in statistics(p<0.05).5.Detection of level of hydroxyproline in hepatic tissue: compared with contol group,the hydroxyproline in hepatic tissue of rat in model group was significantly increased ( P<0.001 ) . compared with model group , the hydroxyproline in hepatic tissue of rat in Complex Prescription group decreased obviously(P<0.01).Conclusion:1. the complex prescription of traditional cheinese medicine could decrease obviously the ALT and AST in serum, increase ALB in serum,hold-back the production of oxidative product MDA,repair the damage of hepatic cell envelope. which indicated the complex prescription of traditional cheinese medicine has curative effect in improving hepatic function .2. the complex prescription of traditional cheinese medicine could decrease obviously the activity of HA,LN,Ⅳ-C,PC-Ⅲin serum, decrease obviously the level of hydroxyproline in hepatic tissue, which indicated the complex prescription of traditional cheinese medicine could hold-back the synthesis of collagen and treat rat hepatic fibrosis .
