Objective To ivestigate the expression of survivin mRNA in the tissue of renal cell carcinoma (RCC) and in peripheral blood preoperatively and postoperatively. To investigate their correlation in differrent pathological grades and clinical stages of RCC. Method 1. The expression of Survivin mRNA in the tissue of 32 cases of RCC and 10 cases of normal kidney was examined by real-time fluorescent quantitative PCR (RFQ-PCR). Each group were analyzed by SPSS11.5 for Windows software.2. The expression of Survivin mRNA in peripheral blood preoperatively,one day postoperatively and one week of postoperatively of 32 cases of RCC and 10 cases of normal kidney was examined by the same method. Each group were analyzed by SPSS 11.5 for Windows software.Results 1. Survivin mRNA protein was significantly expressed in the tissue of RCC. The△CT value in the RCC group was 7.78±0.96,it was markedly higher than that of the normal kidney group, while its△CT value was13.13±1.39;2. In the poorly differentiated group,the differentiation group and the well-differentiated group, The△CT value of Survivin mRNA was differently 3.27±1.42, 7.86±1.43, 8.52±1.27. There was statistically significant difference in different pathological grades and clinical stages. (P <0.05); InⅠperiod group, The△CT value of Survivin mRNA was 10.74±1.27,while inⅡperiod group,Ⅲperiod group,Ⅳperiod group,t The△CT value of Survivin mRNA was differently 6.09±1.43, 7.20±1.72, 3.18±1.07. There was statistically significant difference in each group of different clinical stages. (P <0.05).3. The△CT value of Survivin mRNA in peripheral blood preoperatively and postoperatively of 32 cases of RCC was 6.32±0.96,while the value in normal kidney group was 14.07±2.93. There was statistically significant difference in each group of different clinical stages. (P <0.05).The△CT value of Survivin mRNA in peripheral blood one day postoperatively of 32 cases of RCC was 6.33±0.88. There was no statistically significant difference in each group of different clinical stages. (P>0.05).While the value one week postoperatively was 11.51±1.12.Comparing with the value of preoperatively and one day postoperatively,the value was in decreasing trend. There was statistically significant difference in each group of different clinical stages. (P <0.05).Conclusion 1. The expression of Suvrivin was significantly higher in RCC than in normal kidney tissue,while There was no Survivin mRNA expressed in the tissue of normal kidney group. 2. The expression of Suvrivin in RCC was positively related to the pathological grades,clinical stages.So dececting Survivin mRNA in peripheral blood of RCC patients could be a new method for assistant diagnosis of RCC.we could evalue the malignancies of RCC.3. The expression of Suvrivin in peripheral blood of RCC patients was significantly decreasing after opration.Therefore,Detecting the expression of Suvrivin in peripheral blood of RCC patients could play an important role in diagnose and the forecasting of tumor micro-metastasis and recurrence after operation of initial RCC.
