Object:To study the effect of low dose radiation(LDR)protein kinase Cαand TERT in tumor cell.Methods:Kunming male mice were randomly divided into five groups:direct tumor-beating group(DT group),low dose radiation 1 group(LDR1 group),low dose radiation 2 group(LDR2 group),low dose radiation 3 group(LDR3 group),low dose radiation 4 group(LDR4 group).Having been raised for one week,mice of LDR1-4 groups and DT group were subcutaneously implanted with S180 sarcoma cells in the left hind legs as an experimental in situ animal model.11 days after the implantation,the mice of LDR 1-4 groups were given 75 mGy whole bodyγ-ray radiation.The mice of LDR 1-4 groups were sacrificed after 8,12,24,48 hours.After implantation,DT group were not givenγ-ray radiation and were sacrificed 24 hours after theγ-ray radiation. PKCαand TERT of tumor cell were analyzed by immunohistochemistry.Results:Compared with DT group,the expression of PKCαand TERT were significantly reduced in the LDR2-4 groups(t=2.12-2.43,p<0.05).PKCαwere much lower in LDR2 group than those in LDR3 and LDR4 groups and the difference had statistical significance(t=2.22,p<0.05).The difference of PKCαin LDR3 and LDR4 groups had not statistical significance(p>0.05).Concentration of TERT in LDR2 and LDR4 group were much higher than those in the LDR3 group,but the LDR2 group was lowher than the LDR4 group(t=2.00-2.38,p<0.05).The difference of PKCαand TERT in LDR1 and DT groups had not statistical significance(p>0.05).A significantly inverse correlation was observed between the expression of hTERT and PKCα(r=-0.279,P<0. 05).Conclusion:Low dose radiation of 75 mGy can decrease the production of PKCαand TERT.
