Establishment Of Sprague-dawley Rat Models Of Chronic Nonbacterial Prostatitis And Study On Its Biological Characteristics

AIMTo establish two Sprague-dawley rat models of chronic nonbacterial prostatitis and study its biological characteristics and influence on reproduction.To explore the effect of Cernilton and its mechanism.METHODS1.Model 1:Model ofautoimmune prostatitisAt the 0 and 7th day,male rats were injected with a saline extract of male rat prostate glands(RPG)20mg/ml,40mg/ml,80mg/ml in Freund's complete adjuvant(FCA) intradermally and the Pertussis-Diphtheria-Tetanus vaccine 0.5ml into the abdominal cavity(IP).At 21st day,male rats were injected with saline extract of rat prostate glands 0.Sml.At 35th day,the rats were sacrificed.2.Model2:Modelofchemicalprostatitis1%λ-Carrageenan saline solution 0.1 ml was injected directly in the prostate.At 35th day,the rats were sacrificed.3.The prostate tissue was observed,evaluating whether the models was successful. The concentration of Ca²⁺,Mg²⁺,estradiol(E2),testosterone(T),and dihydrotestosterone(DHT) were determined,observing its biological characteristics.4.After male and female mice mating(1:1),observing the pregnancy rats,the offspring's growth and development,evaluating its effect on reproductive function.5.Cernilton used in the treatment of experimental autoimmune prostatitis(EAP) and Chemical prostatitis in Sprague-dawley rats.To observe its effect and explore its mechanism. RESULTS1.In autoimmune prostatitis model,both intraluminal and interstitial was damaged, congestive,edema,with lymphocytes and monocytes infiltrated,showing obviously chronic inflammation changes.2.In chemical prostatitis model,Prostate interstitial was full of lymphocytes and monocytes,vascular proliferation and congestive obviously,showing obviously chronic inflammation changes.3.In experimental autoimmune prostatitis,compared with control group,WBC increased significantly in the high-dosage group(P≤0.05).Ca²⁺ increased in low-dosage and middle-dosage group,but the difference was not significant(P＞0.05), Ca²⁺ decreased significantly in high-dosage Group(P≤0.05);IgG were significantly higher in low,middle and high-dosage groups(P≤0.01);T increased significantly in low-dosage group(P≤0.05),T also increased in high-dosage group,but the difference was not significant(P＞0.05),DHT was increased in high-dose group,too,but the difference was not significant(P＞0.05)4.In Chemical prostatitis,compared with the control group,dihydrotestosterone(DHT) were significantly lower in the model group(P≤0.01);prostate coefficient significantly lower(P≤0.05);T,Ca²⁺ and Mg²⁺ concentration increased,but the difference was not significant(P＞0.05).5.Reproductive effects in two models:the analysis of sperm motility results showing that between control group and Carrageenan model group A,B,D level were significantly different(P≤0.05),C-level had no significant difference(P≥0.05),at all levels,between control group and immune model group had no significant difference(P≥0.05).Among the three groups,Female pregnancy rate,the number of offspring and the birth sex ratio had no significant difference(P≥0.05).and the death rate also had no significant difference(P≥0.05).Compared with the control group,The pathology of prostate tissue in the model groups had no significant changes.They were smooth, nodules,no congestion and edema,no inflammatory cell invasion.6.To observe the effect of Cernilton,the levels of Ca²⁺ and T significantly increased in the immune model(P≤0.05);the WBC decreased significantly in chemical model(P≤0.05);the pathological changes in the prostate showed that congestion and edema reduced obviously. CONCLUSIONS1.At the 0 and 7th day,80mg/mlRPG mixed with FCA multi-point injected(intracutaneous,IC) with Pertussis-Diphtheria-Tetanus vaccine 0.5ml injected(peritoneal injection IP),At the 21st day,0.5 ml RPG were injected(IP).This was an effective methods for autoimmune prostatitis model in SD rats.2.Chemical prostatitis model can be made by 1%λ-Carrageenan saline solution 0.1 ml injected in the prostate.3.The sperm motility reduced significantly in chemical model,but there was no significant difference on reproduction.4.Cernilton can be used in the treatment of two types of prostatitis,but the mechanisms were different:by increasing Ca²⁺ and T levels in the immune model and by reducing the WBC counts in chemical model.
