Cirsium japonicum DC.,belonging to the Compositae family,is a perennial plant that is widely distributed in north temperate zone in China.The aerial part or root of this plant has been long used to treat hematemesis,hematuria and carbuncle.Modern pharmacology research has elucidated this plant has lots of pharmacological and biological activities such as hemostasia,anti-bacteria and anti-cancer.Since the chemical constituents of Cirsium japonicum DC.are complicated,many active compounds have not been found yet,while the relationship between chemical compounds and clinical uses should be further elucidated.We systematically investigated the chemical constituents of Cirsium japonicum DC..Ten compounds were isolated from the 50%ethanol extract of the whole herb Cirsium japonicum DC..On the basis of their characteristics and spectra,eight structures of the compounds were established as(-)3,4-dihydroxy-2-(4-hydroxy-3-methoxyphenyl)-4-(4-hydroxy-3-methoxybenzyl)-3-tetrahydrofuranmethanol (1),tracheloside(2), linarin(3),pectolinarin(4),hispidulin(5),apigenin(6),caffeic acid(7),and p-coumaric acid(8).Compound 1 and 2 were two lignans,which was first reported that lignan was discovered in the Genus Cirsium and compound 7 was first isolated from this species.Compound 3,4,9 and 10 have been shown the hemostatic activity in vitro.It has been reported that two flavone glycosides,pectolinarin(4) and linarin(3) which have relatively high quantity in Cirsium japonicum DC.,play an important part in hemostatic activity.We have established an isocratic HPLC method for the simultaneous determination of pectolinarin and linarin in Cirsium japonicum DC.. This method is simple,rapid,accurate,and reproducible.It can be an effective method for the determination and quality control of Cirsium japonicum DC..
