Uygur medicine:Cirsium japonicum DC, uyghur: . .It is Compositae thistle dry ground part or root, it is effective in cooling blood and hemostasis, scattered stasis detumescence, reducing fever and causing diuresis. It has extensive medicinal value. This thesis includes two parts for research of Cirsium japonicum DC in xinjiang on the basis of the former achievements .The first part introduces the development of Uygur medicine, the herbal medicine origin, resesrch summary of chemical components, pharmacological effects and clinic- al applications.The second part studied emphatically the extraction and the content determination of total flavonoids, polysaccharides, volatile oil and total alkaloids, and then HPLC- MS analysis, antio-xidation, bacteriostasis and inhibiting nitrosation of the extraction from Cirsium japonicum DC. Besides, the 12 trace elements in the crude drug were determined by ICP-AES.(1) Ultrasonic extraction and reflux extraction of total flavonoids from Cirsium japonicum DC were analyzed and compared, the optimized condition is: 75% ethanol, the rate of solid-liquid 1:60, extract 5 hours, and extraction amount of total flavonoids was 4.14 mg/g. HPLC-MS~n was used to an analyze some compounds from the metha- nol extract of Cirsium japonicum DC, we can identify four of five compounds: Chlorogenic acid, 3,5-Dicaffeoylquinic acid, Rutin and Diosmin. 3,5-Dicaffeoylquinic acid and Diosmin were first found in Cirsium japonicum DC. Also the content of Rutin and Chlorogenic acid were first determined by HPLC.(2) Ultrasonic extraction and hot water extraction were used for polysaccharides from Cirsium japonicum DC, after the conversion factor of polysaccharide to glucose, the content of polysaccharides was measured by phenol-sulfunric acid method. With Berberine Hydrochloride as standard product, the content of total alkaloid was determined by spectrophotometry. (3) The antioxidant activities and inhibiting nitrosation of total flavonoids extract and polysaccharides were evaluated by various antioxidant assays, including aydroxyl radical scavenging, superoxide anion radical scavenging, the results show that they have certain antioxidability, but worse than VC, and polysaccharides have poor effect in superoxide anion radical scavenging. Filter paper and spread-plate method were used to determine their antimicrobial activities and minimum inhibitory concentrat- ion(MIC). Results indicated that the antimicrobial activities of the total flavonoids extract and total alkaloid against five tested microbes were largely different.(4) The volatile constituents was separated and examined by GC-MS, Results showed that 23 kinds of chemical compounds were separated, and 21 kinds of them were identified, some were first found in Cirsium japonicum DC. Digestion with mic- rowave, an analysis method for simultaneously determining more than ten elements by ICP-AES is established.
