Relationship Of The Expression Of INOS And HIF-1α With Angiogenesis In Osteosarcoma

Objective : To study the relationship of the expression of iNOS and HIF-1αwith angiogenesis in osteosarcoma,and to analyize the correlation of both。Methods : The expression of iNOS and HIF-1αprotein were detected by immunohistochemical S-P methods in 64 cases of osteosarcoma and 10 cases of normal bone tissue and 6 cases of osteofibrous dysplasia,CD34 marking vascular endothelial cell was used to measure the microvascular density (MVD)。Western Blot detected the expression of iNOS and HIF-1αin 8 cases of osteosarcoma and normal bone tissue.Results: Immunohistochemical S-P : the poisitive expression rates of iNOS and HIF-1αwere 71.9%(46/64) and 57.8%(37/64) in 64 cases of osteosarcoma, showing a significantly difference in the expression of iNOS and HIF-1αbetween osteosarcoma and control group;The coincidence rate of the expression of iNOS and HIF-1αwas 76.6%(49/64), showing a close relation between them, and the expression of iNOS and HIF-1αwere significantly correlated with stage of the tumor;The medium MVD was 21.43±8.56, much higher in tumor with possitive expression of iNOS and HIF-1αthan in tumor with negative expression of them, there was a significantly difference between them. Western Blot detected the expression of iNOS and HIF-1αin osteosarcoma: the result is approximately coincidence with immunohistochemical S-P.Conclusions: There has a overexpression of iNOS and HIF-1αin osteosarcoma, they have a synergistic effect in new angiogenesis of the tumor, it may play an important role in the genesis,development and metastasis of osteosarcoma。...
