Lotus leaves is a kind of natural plant material used both in food and medicine field. The alkaloid compounds is the main physical activity material in Lotus leaves. Pharmacological studies show that alkaloids in lotus leaf have effect on lipid-lowering diet, anti-virus, anti-bacterial and anti-spasm. This paper study on the determination, extraction, purification technology of alkaloids in Lotus leaves and provided available data for the development and utilization of alkaloids in lotus leaves. The results are as follows:1. Used acid dyes colorimetry to determinate the total alkaloid content in lotus leaves andapplicated "uniform design 3.00 system" software to design the U*10 (108 ) uniform design experiments. This paper has investigated three major impact factors of alkaloid content in lotus leaves, as follows: buffer pH, buffer consumption, bromocresol green consumption. This text has established the best conditions of the acid dyes way determinate total alkaloid content in lotus leaves, which is the pH value of buffer is 2.0, the amount of buffer is 0.5 mL and the amount of bromocresol green is 1.0 mL.2. Used ethanol as solvent and took heating-returning way to extract the total alkaloids of lotus leaf, applicated "uniform design 3.00 system" software to design the U*10(10*53) uniform design experiments. This paper has investigated four major impact factors of extraction effects of alkaloid of lotus leaves, as follows, the temperature of extraction, the concentration of alcohol, extracting time and the ratio of material to liquid. This text has established the best extracting conditions of alkaloid content of lotus leaves,which is used temperature of 80℃, alcohol concentration of 90% ,the extraction time for 4h, the ratio of material to liquid is 1 : 30. The extraction rate is 0.95 percent under used this technology to extracting alkaloids of lotus leaves.3. Used macroporous resin to enrich and purify the rough alkaloids extracted from lotus leaves. Determined D-101 macroporous resin as the best resin through filter from the five macroporous resin. Study on the parameters of adsorption and de-sorption of alkaloids through D-101 macroporous resin has determined the best conditions of used D-101 macroporous resin to purify alkaloids of lotus leaves, which are used the concentration of salt ions with 1 mol/L, pH value of 10 , feed rate of 1.5 BV/h. The best eluting conditions are as follows: used pH 3.0 and the concentration of 70 percent ethanol as eluent, the flow rate was 1.0 BV/h. Used the conditions of this technology could gain the purified total alkaloids products of lotus leaf with purity of 32.48 percent, the purity of nuciferine is 3.37 percent.All in all, this research started with the present problems in performance manufacture technology of alkaloids in lotus leaf, promoted the technological upgrading in quantitative determination, extraction, separation and purification. It also achieved the goal of high per -formance manufacture technology of alkaloids from lotus leaf. It has provided important reference both on theory and technology for high performance manufacture technology and utilization of alkaloids from lotus leaf.
