The leaf of Nelumbo nucifera Gaertn is widespread in the world. Lotus leaf is a kind of natural plant material used both in food and medicine field, involving many active materials. The clinic experience of many years shows that lotus leaf has the function of lipid regulation, and flavonoids and alkaloids are the main active materials, but their effects of lipid regulation were not clear, and the exploiture of lotus leaf was limited. My research focuses on the extraction, isolation, purification and the structure of flavonoids. Then compare the effect of flavonoids and alkaloids on lipid regulation, to open out the functional ingredients of lipid regulation, the results have the value of theory and practice in all-around development of lotus leaf. The main results were as follows:1. The optimum parameters of extracting flavonoidsWith the extraction ratio of flavonoids as studying index, based on the single factor experiment, the optimum parameters and scopes are ensured by the method of orthogonal design L9(34), The results showed that, the ratio of lotus leaf to ethanol, the extraction temperature and the extraction time were the main factors to affect the extraction rate of flavonoids. The optimum parameters are as follows: the concentration of ethanol was 60%, the temperature was 70℃, the time was 40min, the ratio of material to ethanol was 1:40, and the extraction rate of flavonoids is 100.92±0.37mg/g.2. The isolation and purification of flavonoids in lotus leafMacroporous adsorption resin HZ-806 was chosen by experiment of static adsorption-desorption of four kinds of resin HZ-801,HZ-802,HZ-803 and HZ-806 , and the factor of affecting the rate of adsorption and desorption of resin HZ-806 are researched, such as effluent speed, effluent conrentration, volume of eluting agents, elution concentration, in order to find out the optimum conditions, and the results showed that it has good adsorbability and selectivity, the purity of flavonoids was enhanced from 28.97% to 89.04%.
