Guang fangji is the root of Aristolochia fangchi Y. C. Wu ex L. D. Chou et S. M. Hwang. It′s widely used for treatment of rheumatism , pains and elimination quantity of heat ,diuresis. It also has effects in inducing the bite of viper. Guang fang ji mainly grows at Guangdong , Guangxi Province in China. Up to now, people have found Aristolochin acid A, B,and C, Aristololacton,magnolorine,allantoin andβ- sitosterol in the Guang fang ji.For the sake of proving this herb can be used in reason as medicine, this paper described the extraction, isolation, determinations and the content dedermination of representative constituence in the Guang fangchi.Extraction and isolationDried Guang fangchi (3kg) were extracted by heat influx in ethylacetate (15L, 12L, 10L respectively) for three times (2h, 1.5h and 1h, respectively). The extracted solution were amalgamated, decompressed and concentrated to cream, then it (100g) was subjected to silica gel column chromatography with ethylacetate–methanol–water as mobile phase, ODS column chromatography with methanol-water as mobile phase and Sephadex LH-20 column chromatograph with methanol as mobile phase to afford RH1(120m),RH2(9 mg),RH3(18mg),RH4(8mg),RH5(10mg),RH6(10mg),RH7(80mg),RH8(48mg)and RH9(115mg).IdentificationCompounds RH1—RH9 were identified respectively by 1H NMR,13C NMR DEPT,MS,etc. as Isorhamnetin-3-O-robinobioside(RH1), 9-hydroxyl-3,4,8-trimethoxy aristoloctam(RH2), aristoloctam-N-β-D-glucopyanoside(RH3), aristoloctamⅠa N-β-D-glucopyanoside(RH4), 9-hydroxy-aristo-loc-tam-8-O-β-D-rutinoside(RH5), arist-olochicacidⅢa(RH6), daucosterol(RH7),β-sitosterol (RH8) and aristolochic acid-Ⅰ(RH9).But for compound RH8 and RH9, all the compounds were isolated from Guang fangchi for the first time, and RH2 has not been reported before. Quantitative analysis of flavonoids in the Radix Aristolochiae fangchiIn this paper we present a more sensitive quantitative method by HPLC for determination of the main flavonoids RH1 in the Guang fangji.HPLC was performed on a ZORBAX Eclipse XDBC18(250×4.6 mm,5μm) at 30℃with the rate of 1.2 mL/min ,using methanol and 0.1% HAc aq. (0~20min as 47:53,20~30min as (100:0) mobile phase.The content of the Isorhamnetin-3-O-robinobioside in the Guang fangchi pur-chased from different regions were determined by the above method, the results showed that their contents were variety in different regions. The content of Isorhamnetin-3-O-robinobioside extracted from Beijing, Hebei and Anhui province wer-e 0.27%, 0.35% and 0.34% respectively.The results of this paper can offer a new index of content determination for establishment of quality standard of Guang fangji,also can offer science foundation for the development of this medicine plant.
