Hibiscus mutabilis is one of the species in the genus Hibiscus (Malvcaeae), whose driedleaves are used as an important folk medicine for the biological activity of anti-inflammatoryand a good therapeutic effect on skin and cardiovascular diseases. Phytochemical componentsinvestigations of Hibiscus mutabilis leaves have not been previously reported.There are43species in the genus Cicer around the world, and the most well studied andmost vulnerable species to disease is the chickpea(Cicer arietinum). Chickpea is consideredas a kind of precious healthy food because consumption of chickpeas can significantlyincrease the level of blood glucose, blood lipid and blood pressure.The chemical constituents were isolated from Hibiscus mutabilis leaves and chickpea bythe methods of solvent extraction,column chromatography on silica gel,MCI,SephadexLH-20and HPLC, etc. There are19coumpouds (1~19) from the the EtOA fraction ofHibiscus mutabilis leaves,4coumpouds (20~23) from the the EtOA fraction and2coumpouds (24~25) from the n-BuOH-soluble fraction of chickpea.The structures wereelucidated through NMR,MS and established as: kaempferol (1),quercetin (2),quercetin-3-β-D-glucoside (3),kaempferol-3-β-D-glucoside (4),3’-methoxy-quercetin-3-O-glucoside(5),kaempferol-3-O-β-robinobinoside (6),kaempferol-3-O-β-rutinoside (7),quercetin-3-O-β-robinobinoside (8),rutin (9),kaempferol-3-β-D-(6-O-trans-p-coumaroyl)glucopyranosid-e (10),(kaempferol-3-β-D-(6-O-cis-p-coumaroyl)glucopyranoside (11),1,2-benzenedicar-boxylic acid,1,2-bis(2-methylpropyl)ester (12),E-p-coumaric acid (13),Z-p-coumaricacid (14),2-β-D-glucop-yranosyloxy-5-hydroxy-phenylacetic acid (15),daucosterol (16),2,3-dihydrox-2-(4-hydroxy-3-methoxyphenyl)-3-hydroxy-7-methoxybenzofuran-5-carboxylicacid (17),2-dihydrod-ehydrodiconiferyl alcohol (18), pelargonidin-3-O-β-glucopyran-oside (19),β-sitosterol (20),p-hydroxybenzoic acid (21),adenosine (22),1,2-benzen-edicarboxylic acid,1,2-bis(2-meth-ylpropyl) ester (23),1-ethyl-α-L-galactoside (24),(1S,3S)-1-methyl-1,2,3,4-tetrahydro-carboline-3-carboxylic acid (25).We use HPLC to determine the content of four flavonides, namely quercetin-3-O-β-robinobinoside,rutin,kaempferol-3-O-β-rutinoside and kaempferol-3-O-β-ro binobinoside in70%ethanol extract from the leaves of Hibiscus mutabilis simultaneously firstly.The methodis sensitive,accurate and specific,which could provide an important reference to establish thequality control method for this Chinese folk medicine.
