Objective:1.To replicate an available animal model of oral resuscitation in dogs with 40%of blood volume loss.2.To investigate the effects of delayed resuscitation on immune response in dogs subjected to 40%of blood volume loss.3.To investigate the effects of early oral resuscitation on immune response of lymphocyte in dogs subjected to 40%of blood volume loss.Methods:Fourteen dogs were subjected to a 40%hemorrhagic injury from the right common carotid artery.The peripheral venous blood samples were collected at pre-hemorrhage and 1h,4h,8h,24h,48h,72 hours after hemorrhage to analyzed the lymphocyte apoptosis rate,lymphocyte proliferation ability,the percentage of CD3~+/CD4~+ and CD3~+/CD8~+ cells, the ratio of CD4~+/CD~+8 T cells and plasma levels of TNF-α.IFN-γ,IL-2. IL-4 and IL-10.Result:Compared with that in Delayed resuscitation group,the percentage of CD4~+ T lymphocytes were obviously increased in 48h,72h(P<0.05), the percentage of CD8~+ T lymphocytes were obviously decreased in 24h. 48h.and 72h(P<0.01),and the ratio of CD4/CD8 T cells were increased dramatically in 24h.48h,and 72h(P<0.01 ) alter hemorrhage shock in Oral resuscitation group.Lymphocyte apoptosis rate were increased at 4h,48h and 72h(P<0.05,P<0.05 and P<0.01 respectively),and lymphocyte proliferation ability was elevated at 24h after hemorrhage shock in Oral resuscitation group.Plasma levels of IFN-γwas elevated at 8h,24h(P<0.01).IL -4 were decreased at 1h.24h,and 48h(P<0.05 ) and the ratio of IFN-γ/IL-4 were elevated at 1h.4h,24h and 72h(P<0.05,P<0.05,P<0.01 and P<0.05 respectively) in animals resuscitation with oral GES compared with delayed resuscitation.IL-10 concentration were elevated at 1h,4h and 72h(P<0.05 ).TNF-αconcentration were decreased at 24h,48h and 72h(P<0.05,P<0.01 and P<0.05.respectively) in oral-resuscitation dogs compared with delayed-resuscitation dogs.At 1h,4h,and 8h(P<0.05) the circulating concentration of IL-2 was increased in oral-resuscitation dogs compared with delayed-resuscitation dogs.Conclusion:Immune response of dogs subjected to 40%of blood volume loss was depressed.Delayed resuscitation had no significant beneficial effects on immune function in this model.And early Oral resuscitation with GES had obviously immune benefit compared with delayed resuscitation.
