LRP16 is a human gene cloned from the peripheral blood lymphocyte of the healthy adult and overexpressed in the malignant tumor depended on estrogen.In recent years,the metabolism of tumor tissue becomes a research hotspot.Some oncogenes take part not only in promoting proliferation,inhibiting apoptosis,but also in changing the mode of the metabolism of tumor tissues to adapt them to survive in the environment,which is hypoxia and lack of nutrient supply.This suggests that oncogenes may also affect the body's metabolism—which have some evidences.And some studies have found that some obesity-related genes are involved in tumorigenesis.So we suppose that the tumor and obesity may have a "common origin",that is,the excessive activation(or inhibition) of certain genes may lead to tumorigenesis and obesity.LRP16 gene,as an oncogene and an estrogen-related gene, may have an impact on body's metabolism.The present study shows LRP16 plays a role in adipocyte differentiation,insulin resistance,etc.The study can be divided into the following three parts:1.Analysis of effect of LRP16 gene on the mature adipocytes with cDNA microarrayObjective:To investigate LRP16 gene's functions in the mature adipocytes and it's potential clinical significance.Methods:In this study,high-flux microarray technology was employed to analyze the genes differentially expressed between the experimental group which overexpressed LRP16 and the control group.Public database was also used to analyze the probable signaling pathways and target genes which LRP16 may be involved in.Results:The number of genes differentially expressed between two groups is 1,222,of which 653 up,569 down.The involved signaling pathways are insulin resistance,glucose and lipid metabolism,adipocyte differentiation and so on.Conclusion:LRP16's function is to promote the(Micro) inflammation,and it may has an impact on adipocyte differentiation,insulin resistance, glucose and lipid metabolism.2.LRP16 may promote a fat burning-Like phenotype in White AdipocytesObjective and Methods:In this study,we investigated possible remodeling effects of LRP16 in 3T3-L1 adipocytes.Changes in the morphous and expression of genes related to thermogenesis were observed.Results:The overexpressed LRP16 triggered an up-regulation of the mRNA levels of peroxisome proliferator-activated receptorγcoactivator-1α(PGC-1α),uncoupling proteins 2 and muscle-type carnitine palmitoyltransferase 1(CPT1).Conclusions:The results indicate that LRP16 favors the acquisition of brown adipocytes-like properties in 3T3-L1 adipocytes. Understanding the mechanisms and effectors involved in the remodeling of 3T3-L1 adipocytes can contribute to new avenues of prevention and treatment of obesity and type 2 diabetes.3.LRP16 promotes the expression of adiponectin and leptinObjective and Methods:In this study,we investigated effects of LRP16 on adipokines in 3T3-L1 adipocytes.Changes in the mRNA levels of adiponectin and leptin were observed.Results:The overexpressed LRP16 triggered an up-regulation of the mRNA levels of adiponectin and leptin.Conclusions:LRP16 may promote energy expenditure and improve insulin resistance.Conclusions1.LRP16's function is to promote the(Micro) inflammation,and it may has an impact on adipocyte differentiation,insulin resistance,glucose and lipid metabolism.2.LRP16 favors the acquisition of brown adipocytes-like properties in 3T3-L1 adipocytes.3.LRP16 may promote energy expenditure and improve insulin resistance.
