【Objective】To build several different models of the bile duct injury of pigs repaired with acellular dermal matrix,and observe the influence of gallbladder fistulation tube on regeneration of bile duct cells and liver pathological changes in different groups,and observe the regeneration of bile duct cells and liver pathological changes in different groups.To explore the application of gallbladder fistulation tube in the experiment on pigs which bile duct injuries were repaired with ADM.【Methods】24 healthy Dian-Nan small-ear pigs in closed group were divided randomly into four groups of male and female.Group SC was the repairation for a segmental bile duct injury with ADM patch and cholecystostomy group(n=6);Group SI was the repairation for a segmental bile duct injury with ADM patch surgery group (n=6);Group PC was the repairation for partial bile duct injury with ADM patch and cholecystostomy group(n=6);Group PI was the repairation for partial bile duct injury with ADM patch surgeD,group(n=6);The animals' postoperative performance status were evaluated.MRCP was used to define the patency of bile ducts at 12.48weeks.The animals' postoperative blood routine investigation and changes in liver function were examinated at 1,3,12,24,48weeks.Collected specimens at 6.12.24,48 weeks after the operation,then observed regeneration of the tissues of bile duct and the pathological changes of livers.【Results】Group SC and group PC were surgical success.MRCP,blood routine and liver function were examinated at regular time and the most were showed abnormal. The regeneration of bile duct epidermis was not good and the hyperplasia of fiber organization was active at 6,12weeks,and bile duct stricture,liver fibrosis at 24weeks. and liver cirrhosis at 48weeks.Group SI and group PI were surgical success.MRCP, blood routine and liver function were examinated at 12,48weeks and the most were normal.Tissues of bile ducts were taken for examination to observe the regeneration process and found them in upward trend at 6,12-weeks,return to normal at 24-weeks, and no change at 48weeks.There was also no obvious pathological changes in liver.【Conclusions】(1) The models of pigs which bile duct injuries were repaired with acellular dermal matrix patch were successful.(2) There was not obvious complication in group SI and group PI for a long time after operation.(3) There were many complicating diseases in group SC and group PC after 12weeks.(4) Perhaps it was the gallbladder fistulation tube that induced inflammation and cholestasis which led to the high-level expression of TGF-β1,then bile duct stricture and liver cirrhosis. (5) The gallbladder fistulation tube had an adverse effect on the experiment.
