Bankground&Objective:Thyroid cancer is the most common malignant tumor in endocrine system.Its incidence rate rises year by year.The research about relationship between tumor and matrix metalloproteinases(MMPs),matrix metalloproteinase inhibitors(TIMPs) is one of the hot spots in modern cancer research.This experiment was designed to study matrix metalloproteinase MMP-1,MMP-7 and tissue inhibitor of matrix metalioproteinase TIMP-1,TIMP-2 in thyroid carcinoma and its invasion and metastasis to explore the occurrence of thyroid cancer's development,in order to clarify the pathogenesis of thvroid cancer and explore new methods to provide some Oasis for new prevention and treatment.Methods:The expression and distribution of MMP-1mRNA,MMP-7 mRNA, TIMP-1mRNA,TIMP-2mRNA was detected by in-situ hybridization in 60 cases of thyroid cancer specimens(35 cases of papillary carcinoma,10 cases of medullary carcinoma.8 cases of follicular carcinoma,7 cases of undifferentiated carcinoma),12 cases of thyroid adenoma specimens and 26 cases of normal thyroid tissue as control, and the expression levels of TIMP-1 protein and TIMP-2 protein was also detected using immunohistochemical SP method in these organizations.Results:The experession rates of TIMP-1mRNA and protein in thyroid cancer, thyroid adenoma and normal thyroid tissues were:58.3%,63.3%;50.0%,41.7%; 15.4%,23.1%respectively.The experession rates of TIMP-2mRNA and protein in thyroid carcinoma were 65.0%,68.3%,and in thyroid adenoma were 50.0%,50.0%, in normal thyroid tissue were 7.7%,7.7%respectively;The expression of TIMP-1mRNA and protein,TIMP-1 mRNA and protein was no correlated to the lymph node metastasis and invasion of tumor(P>0.05).But TIMP-1mRNA and TIMP-2mRNA was correlated to Clinical histological types(P<0.01).Compared with normal thyroid tissue,the hybridization signal of MMP-1mRNA,MMP-7mRNA of kinds of tumor tissues has a rising tendency,and compare various types of thyroid cancer,the hybridization signal of MMP-1mRNA,MMP-7mRNA in hyroid adenoma tissue was not weakening obviously.At the same time,there wasn't significantly positive modulating correlation between the expression of MMP-1mRNA,MMP-7 mRNA and clinopathological factors(Clinical stage,histological type,gender,age, lymph node metastasis)(P>0.05)。Conclusion:1.TIMP-1mRNA and protein,TIMP-2mRNA and protein,MMP-1mRNA and MMP-7mRNA in thvroid cancer are highly expressed.2.TIMP-1mRNA and protein,TIMP-2mRNA and protein,MMP-1mRNA and MMP-7mRNA are not closely related with the Clinical stage,histological type,gender, age and lymphnode metastasis.but the expression of TIMP-1mRNA and TIMP-2mRNA are related to Clinical histological types.3.TIMP-1mRNA and protein.TIMP-2mRNA and protein.MMP-1mRNA and MMP-7mRNA are valuable clinical diagnosis references.
