Objectives:To investigate the effects of CUMS on platelet aggregation,platelet 5-HT and histopathological change of coronary artery and aorta in rat.Method:In the first two weeks,48 male SD rats were randomly and averagely divided into 2 groups.The rats in HFD groups and controls group were fed with fat diet and basic food respectively.The rats in HFD groups were injected with VitD3 6000001U/kg Intraperitoneally once,and then fed with Propylthiouracil by intragastric administration every day.The rats in CT groups were Only intraperitoneally injected with Sodium Chloride, and then fed with Sodium Chloride by intragastric administration every day.In the next seven weeks,the rats in HFD groups were randomly and averagely divided into HFD' group(n=22) and HFD+Stress group(HFDS group)(n=12).The rats in CT groups were randomly and averagely divided into CT' group(n=12) and CT+Stress group(CTS group) (n=22).The rats in HFDS and CTS groups were given CUMS.The rats were weighed every end of week.At the end of the 9th week,all rats were executed after the blood platelet aggregation,platelet 5-HT was collected and Histopathological change of coronary artery and aorta were appreciated.Results:1.Body weight of the rats were tested by one-way ANOVA.Compared with CT' group, body weight of the rats both in CTS and HFDS groups at the end of 5th week decreased significantly at the end of 5th week(P<0.05),Body weight of the rats in CTS group was compared with CT' group's decreased significantly at the end of 6th week(P<0.05),while Body weight of the rats in HFD' group compared with CT' group's increased significantly at the end of 7th,8th,9th weeks(P<0.05).2.The platelet aggregation of rats was 36.4±14.8%,43.8±16.8%,54.0±22.7%,65.8±25.5%in CT' group,CTS group,HFD' group, HFDS group respectively.Tested by one-way ANOVA,Compared with CT' group,platelet aggregation of rats in HFDS group increased significantly(P<0.05).Tested by post hoc analysis,HFD has significant effect on increasing platelet aggregation(P<0.05) while CUMS has no significant effect on increasing platelet aggregation(P>0.05),but there was no effect of interaction between HFD and CUMS to platelet aggregation(P>0.05). 3.The platelet 5-HT of the rats was 0.272±0.332 umol/109 plt,0.137±0.184umol/109 plt, 0.502±0.409umol/109 plt,0.187±0.216umol/109 plt in CT' group,CTS group,HFD' group, HFDS group respectively.Tested by one-way ANOVA,there was no significant difference between every group(P>0.05).Tested by post hoc analysis,HFD has no significant effect on platelet 5-HT(P>0.05),CUMS has significant effect on decreasing platelet 5-HT(P<0.05),there was no effect of interaction between HFD and CUMS on platelet 5-HT(P>0.05).4.the scores of histopathological change of coronary artery and aorta of the rats were 0.22±0.16,0.61±0.52,1.19±0.48,1.50±0.44 in CT' group,CTS group,HFD' group,HFDS group respectively.Tested by post hoc an- alysis,HFD has significant effect on score of histopathological change of coronary artery and aorta(P<0.05),CUMS has significant effect on the score of histopathological change of coronary artery and aorta (P<0.05),there was no effect of interaction between HFD and CUMS on histopathological change of coronary artery and aorta(P>0.05).Conclusion:Body weight of the rats decreased in early of CUMS,while increased again with time passing by.HFD has effect on increasing Body weight of the rats quickly.CUMS has no effect on platelet aggregation of the rats.HFD has effect on increasing platelet aggregation of the rats.there was no synergy between HFD and CUMS to platelet aggregation. HFD has no effect on platelet 5-HT of the rats.CUMS has effect on decreasing platelet 5-HT of the rats.there was no effect of interaction between HFD and CUMS on platelet 5-HT of the rats.CUMS and HFD could promote atherosclerosis of coronary artery and aorta,there was no synergy between HFD and CUMS to AS of coronary artery and aorta of the rats.
